Objectives: To investigate association of nm23-Hl with sensitivity to chemotherapeutic agents in mice lung carcinoma LA-795, and suppression of nm23-Hl gene on metastasis of LA-795 cell with high metastasis potential. Methods: In this study recombinant eukaryotic expression vector pcDNA3-nm23-Hl containing human nm23-Hl cDNA and vector pcDNAS are transfected by lipofectamine regent into mice lung carcinoma cell LA-795. The positive transfectant is obtained by using G418 screening. Western-blot shows that Nm23-Hl/NDPK-A is steadily expressed in the positive transfectant. Anti-metastasis of nm23-Hl is demonstrated by agarose drop, soft agarose clone and monolayer invasion assay. Sensitivity associated with chemotherapeutic agents including cisplatin, mechlorethamine, mitomycin C, 5-fluorouracil, vincristine and vp-16 are determined by MTT assay and the same to inhibition of nm23-Hl on cell growth.Results: (1) nm23-H1 is associated with sensitivity to cisplatin , mechlorethamine, mitomycin C but not 5-fluorouracil, vincristine and vp-16; (2) nm23-Hl don't indicate prohibition on gowth of LA-795 cells. (3) nm23-Hl suppresses greatly metastasis potential of LA-795 cells.Conclusions: nm23-Hl can suppress metastasis potential of LA-795 cells and enhance sensitivity of LA-795 to alkylating agents. |
