Lipometabolism disorder plays an important role in atherosclerotic development, the danger of coronary heart disease (CHD) will decrease 2-3% if serum total cholesterol(TC)decrease 1%. So the lipid-regulation drugs which can treat Lipometabolism disorder have been used to prevent and treat atherosclerosis(AS) and CHD. XIAO YU JIAO NANG (XYJN) Capsule is made of the effective ingredients of Crataegus pinnatifida Bge and salvia miltiorrhiza Bge which combine togather with special ratio. In our stady, the effects of different does of XYJN on rabbit hyperlipidemia, hemorrheology and anti-oxidation were observed, the clinical effecf of the drug on 20 cases of primary hyperlipidemia was studied as will.1. The effecf of XYJN on blood lipidNew Zealand rabbit were fed with high-fat diet and/or administrating with XYJN stimultaneously at dose of 25mg/kg.d. 50mg/kg.d.100mg/kg.d. After 2 weeks of treatment, serum TC was decreased by 12.6%-34.1%, espe-cially in the high dose group, the action was more effective as compare with the hyperlipidemia group (p<0.05). Serum TG, LDL-C and ratio of TC/HDL-C were decreased by 25.0-45.4%, 1.2-30.4% and 14.1-30. l%,respectively, but there is no significant statistical difference. While serum apolipoproteinB(ApoB) was decreased significantly(p<0.05-0.01). After 4 weeks of treatmeat, serum TC and LDL-C were decreased significantly compare with the hyperlipidemia group(p<0.01),and with apparent dose dependancy. Serum TG and ApoB were decreased by 13.2%-43.6% and 6.5%-32.0%. respectively(p<0.05-0.01). XYJN can not elevate the HDL-C, but can decrease the ratio of TC/HDL-C significantly (P<0.01).It can be conferred that XYJN can prevent the rabbits from hyperlipidemia.2. Effect of XYJN on atherosclerosisAfter 4 weeks of high-fat food and treatment of XYJN, the foma cells of abdominal artery obviously diminished in New Zealand ribbit. As can be prevented in drug groups.3. Effect of XYJN on liver fat and liver weight indexAfter 4 week of high-fat food and treatment of XYJN ,the deposition of TC and TG can be prevented in New Zealand rabbit liver, Liver weight coeffective and hepatic fatty of the groups with drug administrating was lower than the hyperlipidemia group(P<0.01).4. Effect of XYJN on platelet aggregationAfter 4 week of high-fat food and treatment of XYJN ,XYJN can inhibit the platelet aggregation induced by ADP and collagen, and the high-dose drug can inhibit significantly the platelet aggregation induced by collagen(P<0.05)5. Effect of XYJN on hemorrheologyAfter 4 week of high-fat food and treatment of XYJN ,the blood viscosity,blood reduction viscosity,red cell rigidity index and deformation index of the rabbit have the tendancy of decrease. XYJN in high dose group have significant effect on the decrease of blood reduction viscosity, red cell rigidity index and deformation index(P<0.01).It was suggest that the drug was benefit for improving the hemorrheology of the rabbits.6. Effect of XYJN on anti-oxidation and fibrinolysis systemAfter 4 week of high-fat food and treatment of XYJN .the levels of rabbit serum t-PA, SOD were highter and lower serum MDA levels in XYJN group than that in hyperlipidemia group(P<0.01).The results revealed that XYJN possessed the ability of anti-oxidation and can improved the fibrinolysis system.7. Clinical Observation of XYJNXYJN are ustd in treatment of 20 cases of primary hyperlipidemia. At the end of 8 weoks treatment, serum TC, LDL-C and TG were decreased by 33.78%,35.67%and 30.74%, respectively.. Total effectiveness is 95 %, But HDL-C does not increase significantly(P>0.05), In the period of treatment , there is no change in liver, renal funcfion and blood glucose, So it was concluded that XYJN is a effective and safe lipid-regulation drug.Supervisors: Prof. Gu Zhen Lun Prof. Xie Mei Lin Postgraduate: Sun kang Yue...
