Background: Nitric oxide is an inorganic small molecule and it is the most vasodilator. In 1997, Amgrimsson R, et al, suggested that endothelial nitric oxide synthase gene (eNOS gene) could be a familial pregnancy-induced hypertension (PIH) gene.Objective: The aim of this experiment is to identify association between intron 4a/b polymorphism and intron 11 G174A polymorphism of eNOS gene in Chinese and PIH.Methods: Genome DNA from leukocyte was extracted as PCR templet. Using PCR-PAGE and PCR-RFLP to document the polymorphisms of 4a/b and G174A of eNOS gene.Result:1. Detecting the site of 4a/b in intron 4 of eNOS gene4a/b polymorphism was found in Chinese and a new allele 4c was confirmed firstly. The allele distribution is following: The frequency of 4a allele is 9.8%, 4b allele is 89.1% and 4c allele is 1.1% in random women controls. The frequency of 4a allele is 10.5%, 4b is 88.0% and 4c is 1.5% in normal pregnancy women. The frequency of 4a is 13.4%, 4b is 84.8% and 4c is 1.8% in the women with PIH.2. Detecting the site of Gl 74A in intron 11 of eNOS geneThe research group has been divided in three groups. The frequency of G allele is58.9% and A allele is 41.1% in random women controls. The frequency of G allele is 60.6% and A allele is 39.4% in normal pregnancy women. The frequency of G allelel is 56.3% and A allele is 43.7% in the women with PIH.Conclusion1. There is 4a/b polymorphism in intron 4 of eNOS gene in Chinese, and a new allele 4c was confirmed firstly.2. No statistically difference has been found between the normal controls and the women with PIH. It shows that there is not associated between the 4a/b/c polymorphism of eNOS gene and PIH.3. There is G174A polymorphism in intron 11 of eNOS gene in Chinese.4. No statistically difference has been found between the normal controls and the women with PIH. It shows that there is no association between the G174A polymorphism of eNOS gene and PIH.
