ObjectiveTo study the relationship between single nucleotide polymorphisms in the e NOS gene, human cytomegalovirus(HCMV)infection and essential hypertension(EH) in the Han population in Sichuan and investigate the genetic susceptibility to EH in Sichuan. Materials and MethodsA total of 226 patients with EH and 186 healthy subjects we rerecruted in this case-contro l study. The G894 T,T786C and 27 bp VNTR polymorphisms of the e NOS gene we redetected by PCR-RFLP. We compared genotype frequencies and allele gene frequencies between EH group and control group. We also analysed haplotype;at same time,HCMV-DNA were simultaneously tested by nested PCR using two primer sequences from HCMV IEA genome from 412 cases.(EH case:226,healthy control:186). Results(1)Han population increasing obesity,overweight, creatinine, impaired fasting glucose,sare the main risk factors of EH, apoprotein A1 may be the protective factor.(2)By age, sex, weight index of the ratio and Logistic regression analysis of the score, e NOS gene G894 T loci GT+TT genotypes and 27 bp VNTR loci 4a/b genotypes may be the genetic susceptibility in Han population EH of Sichuan, T alleles and 4a/b alleles may be the predispositions in Han population EH of Sichuan. 27 bp VNTR loci 4a/b genotypes may be associated with male EH in Sichuan,G894 T loci T alleles and 27 bp VNTR loci 4a alleles may be the hypertension susceptibility gene for male in Sichuan Han population.Female EH may be associated with 27 bp VNTR loci 4a/b genotypes.The polymorphism of e NOS gene T786 c loci may be not associated with Han population EH in Sichuan(P>0.05).(3)There is a paired strong linkage disequilibrium between e NOS gene G894 T loci and T786 C loci in Sichuan Han population EH and female.(4)C-4b-G may be the risk haplotype for Han population EH and male EH in Sichuan; T-4b-G may has a protective effect against EH in Sichuan Han population(P<0.05);Maybe the haplotype of C-4a-G increasing the risk of female EH in Sichuan Han population;there is a significant difference in haplotype of T-4a-G between male and female,male EH and female EH( P<0.05).(5)There is no significant association between HCMV infection and EH in Sichuan Han populaiton.Increaing serum total protein is associated with HCMV infection in male having EH of Sichuan Han population.The abnormal BMI is associated with HCMV infection in Sichuan EH female. Conclusion(1) e NOS gene G894 T loci GT+TT genotypes and 27 bp VNTR loci 4a/b genotypes may be the genetic susceptibility in Han population EH of Sichuan, T alleles and 4a/b alleles may be the predispositions in Han population EH of Sichuan. 27 bp VNTR loci 4a/b genotypes may be associated with male EH in Sichuan,G894 T loci T alleles and 27 bp VNTR loci 4a alleles may be the hypertension susceptibility gene for male in Sichuan Han population.Female EH may be associated with 27 bp VNTR loci 4a/b genotypes.The polymorphism of e NOS gene T786 c loci may be not associated with Han population EH in Sichuan.(2)There is a paired strong linkage disequilibrium between e NOS gene G894 T loci and T786 C loci in Sichuan Han population(4)C-4b-G may be the risk haplotype for Han population EH; T-4b-G may has a protective effect against EH in Sichuan Han population;(3)There is no significant association between HCMV infection and EH in Sichuan Han populaiton.
