Objective: In this study the apoptosis and expression of caspase-3 protein of neuronal cell surrounding intracerebral hemorrhage were observed to investigate the mechanism of neuronal injury in intracerebral hemorrhage, and investigate the relationship between apoptosis and caspase-3 protein following intracerebral hemorrhage in rats.Methods: Intracerebral hemorrhage was produced in adult rats by double blood injection method, and 60ul autologous blood or paraffin oil was injected into right basal ganglia with the use of sterile technique. The animals' brains were removed at 4 6 hours or at 1, 3, 5, or 10days after hemorrhage. Terminal deoxynucleotidy transferase-mediatd deoxyuridine triphosphate biotim in situ nick end-labeling (TUNEL) was used to detect deoxyribonucleic acid fragmentation. Caspase-3 protein was measured by immunohistochemistry.Result: After the autologous blood was injected , the left body of the rats were paralytic,the activity of the rats decreased and the rats tended to rotated to left. The hematomas could be seen in right basal ganglia of rats. All the hematomas almost had the same size and form. At 6h apoptical cells appeared in the intrcerebral hemorrhage model and were present for more than 10 days, peaking at 3 days. The apoptical cells were concentrated in the area where the pathological change was obvious, but the apoptical cells also could be seen in the area where had no pathological change. But only a few apoptical cellswere found in the paraffin oil control brains. The Caspase-3 protein began to express at 6h after hemorrhage, peaked at 3 days, and then declined gradually.Conclusion:(1)The model of intracerebral hemorrhage in rats has the advantages of easy operation, good repetition and et aI.(2) Apoptosis is a pattern of neuronal cell death after intracerebral hemorrhage. The apoptical neuronal cells appeared at 6h,and peaked at 3d after hemorrhage, and it demonstrated that the time of hematoma evacuation should be in 6hours.(3)The time-phase pattern of Caspase-3 protein is similar to that of neuronal apoptosis. Caspase-3 protein play an important role in the process of apoptosis of neuron after intracerebral hemorrhage. (4)The injury rang ofneuronal cells surrounding the intracerebral hemorrhage was more extensive in molecular level than what we observed with eye or microscope .
