Research On The Influence Of MCAO Rats' Cerebral Function Recovery And The Expression Of EcNOS And P-GP By Acupuncture

Our previous studies have indicated acupuncture can improve the cerebral ischemic rats' microcirculation and accelerate the recovery of cerebral function.To investigate the mechanism of acupuncture on cerebral ischemic vascular diseases,we have studied the expression of endothelial functional corelated genes in the rats' cerebral microangium .The MCAO(middle cerebral artery occlusion) rat model are made by nylon thread embolus and are divided different groups before acupuncture.Renzhong Neigua,Quchi Zusanli,Diji Jingqu are choosen for the treatment of different groups in the chronic ischemic stage after MCAO.We observe the variety of rats' nerve signs and common situation,detect nitric oxide(NO) endothelon(ET) super-oxide dismutase(SOD) MDA glutathione peroxidase(GSH-PX) of blood ,make HE tissue slices to observe the change of rats cerebral cellular morphosis . The expression of endothelial cell nitric oxide synthase(ecNOS) andP-glucoprotein(P-GP) mRNA are also dedected in situ hybridization.Theresults indicates acupuncture can accelerate the disappearance of pathological nerve sign during recovery stage of rats' chronic cerebral ischemia .Quchi Zusanli and Renzhong Neiguan groups have showed the obvious effect among the groups. Acupuncture can decline the amount of NO and MDA,but have not obvious effect on the level of ET\ GSH-PX and SOD in the blood . Acupuncture can decrease the rate of neuronal cell death in the CA3 region of the hippocampus and increase the microangium quantity there. Acupuncture has not obvious influence on the expression ecNOS mRNA in the endothelial cell in the CAS region of the hippocampus in the chronic stage after MCAO,but can increase the express of ecNOS mRNA in neuron. The expression of P-GP mRNA is very weak in the cerebral micrangium cell after MCAO.The effect of P-GP after acupuncture needs further study.
