Objective: To investigate the expression and significance of cyclinE, P27 and Ki-67 in human colorectal carcinoma.Methods: Using S-P immunohistochemistry method, we examined the expression of cyclinE, P27 and Ki-67 in the specimens of 64 cases with colorectal carcinoma and 12 cases with normal mocosa.Results: The positive immunostaining rate of cyclinE was significantly higher in the colorectal carcinoma (57.18%) than in the normal mucosa (16.67%) (P<0.05).The expression of cyclinE correlated with Dukes' stage and lymphocytic infiltration.The cyclinE positive rate in stages Dukes A, Dukes B, Dukes C and Dukes D was 85.00%,55.56%,42.86% and 33.33% respectively, the incidence of cyclinE expression was significantly higher in stage Dukes A than in stages Dukes B, Dukes C and Dukes D. The cyclinE positive rate was higher in these cases with negative lymphocyticinfiltration(71.05%) than those with positive lymphocytic infiltration (38.48%)(P<0.05). cyclinE expression did not correlate with prognosis for patients with colorectal carcinoma. The P27 positive rate was significantly lower in the colorectal carcinoma (53.13%) than in the normal mucosa (91.67%)(P<0.05). The P27 positive rate in stages Dukes A, Dukes B, Dukes C and Dukes D was 75.00%, 66.67%, 28.57% and 25.00% respectively, the expression of P27 in stages Dukes A and Dukes B was higher than in stages Dukes C and Dukes D (P<0.05). The P27 positive rate was lower in these cases with positive lymphocytic infiltration(26.92%) than those with negative lymphocytic infiltration(71.05%)(P<0.05). The 3 or 5-year survival rate was significantly lower in the patients with a tumor of negative P27 expression (46.15%) than in the patients with a tumor of positive P27 expression (73.33%)(P<0.05). The Ki-67 positive rate was significantly higher in cases with colorectal carcinoma (85.94%) than in the normal mucosa (16.67%)(P<0.05). No significant correlation was found between Ki-67 expression and tumor localization, Dukes' stage or patient's prognosis. cyclinE expression correlated with expression of Ki-67. P27 expression did not correlate with expression of cyclinE or Ki-67.Conclusion: The overexpression of cyclinE and down or negative-expression of P27 might accelerate the progression of the cell cycle and promote the carcinogenesis of the colorectal intestine. Theoverexpression of cyclinE might be an relatively earlier event in the initiation of colorectal carcinoma. The P27 down or negative-expression not merely might be associated with other mechanisms responsible for aggressive and metastatic tumor behaviour in colorectal carcinoma, but predicts unfavorable prognosis of colorectal carcinoma. Analysis of P27 expression in colorectal carcinoma is useful for us to estimate the malignance of tumor and predict the patient's prognosis.
