Objective:To explore the expression of CyclinE,MMP-9,Survivin,PTEN and to discuss the correlation between them and the clinicopathological characteristic of colorectal carcinoma. Therefor we can more exactly decide the prognosis of colorectalcarcinoma in molecular level,and provide theory basis for the corresponding gene therapy method.Methods:Immunohistochemistry Envision two-step system was used to detect the expression level of CyclinE,MMP-9,Survivin and PTEN in sixty-six colorectal cancer samples.Then to research relationship between the expression of them and patients'age,gender,tumor location, type,infiltration depth,differentiation degree,Dukes installment and lymph node metastasis status and then to do a statistical analysis.Twelve of normal colorectal tissue as a control,and to do a statistical analysis using the same method.Results:1.In the twelve cases of normal colorectal tissue,the positive expression of CyclinE was one cases,the positive expression of MMP-9 was three cases,all the Survivin was negative expression,but the positive expression of PTEN was l lcases.2.The positive expression rates of CyclinE,MMP-9,Survivin in 66 case of colorectal neoplasms is 62.12%,75.76%,71.21%,that is significantly higher than in coloretal mucosa(8.33%,25.0%,0).The positive expression rates of PTEN in colorectal neoplasms(53.03%)is significantly lower than in normal coloretal mucosa(91.67%,P<0.05).3.CyclinE abnormal expression of colorectal neoplasms was correlated with Dukes stage and lymph node metastasis (P<0.05),MMP-9 expression of colorectal neoplasms was correlated with depth of infiltration,Dukes stage and lymph node metastasis(P<0.05),Survivin expression of colorectal neoplasms was markedly correlated with Dukes stage and lymph node metastasis(P<0.05),PTEN expression of colorectal neoplasms was markedly correlated with depth of infiltration,Dukes stage and lymph node metastasis(P<0.05).4.Correlation analysis showed that the expression of PTEN has a negative correlation with the expression of CyclinE,MMP-9 and survivin,it showed that the role of PTEN and CyclinE, MMP-9,Survivin in colorectal cancer genesis and development is opposition.The expression of MMP-9 was increased with the increased intensity of survivin(r=0.447).Conclusion:1.The expression of CyclinE,MMP-9 and survivin were increased significantly but the expression of PTEN was decreased significantly,it showed that they play an important role in the pathogenesis and development of colorectal carcinoma.2.The expression of CyclinE,MMP-9,Survivin and PTEN have correlation with some clinicopathological characteristics,they expression levels can serve as a useful indicator to detect the degree of malignancy about colorectal cancer.3.The expression of PTEN has a negative correlation with the expression of CyclinE,MMP-9 and Survivin,the expression of PTEN was decreased but the expression of CyclinE,MMP-9 and Survivin were increased,they maybe play a collaborative in the pathogenesis and development of colorectal carcinoma.The expression of MMP-9 has a positive correlation with the expression of Survivin,They may exist together,or each other regulatory echanismIn inhibiting apoptosis and promote tumor angiogenesis,and through different channels to promote the occurrence and development of colorectal cancer.4.The joint detection of these four genes proteinum has a significance value in evaluating biological behaviour in colorectal neoplasms,which maybe a New molecular target therapy site of colorectal carcinoma.
