ObjectiveDetecting the expressions of Livin, Caspase-9protein and the cellularapoptosis in human gastric carcinoma (GC) tissues to explore the relationshipbetween the expressions of Livin, Caspase-9protein and cellular apoptosis.Then explore their effects and possible mechanism during the occurrence anddevelopmen of human gastric carcinoma.MethodsApplying TdT-mediated dUTP-digoxygenin end-labeling (Tunel) to detectthe cellular apoptosis index (AI) in78specimens of gastric carcinoma, thecorresponding tumar-adjacent tissues, nomar tissues, and evaluating thedifferences in three groups. The expressions of Livin and Caspase-9proteinwere detected by immunohistochemical technique and Western blot. And thedifferences of their expressions in three groups were compared, and thecorrelations of their expressions with various clinicopathologic characteristicsand cellular apoptosis in gastric carcinoma were analyzed statistically.ResultsAI in gastric carcimoma tissues was significantly lower than thecorresponding tumar-adjacent tissues and nomar tissues (P<0.01). There wereexpressions of Livin and Caspase-9protein in gastric carcimoma tissues, corresponding tumar-adjacent tissues and normal tissues. The positiveexpression rates of Livin in gastric carcinoma tissues and correspondingtumar-adjacent tissues were significantly higher than that in nomar tissues, andthe expression of Caspase-9was lower than that in normal tissues(P<0.01).The expression of Livin was negatively correlated with the expression ofCaspase-9(r=-0.30,P<0.01). The expression of Livin and Caspase-9wasclosely correlated with tumor depth of gastric cancer, lymph nods metastasis,clinical stages and histological differentiation grade, but not correlated with sex,age and tumor size (P>0.05). And in gastric carcinoma tissues the expression ofLivin was negatively correlated with the AI (r=-0.61, P<0.01), but theexpression of Caspase-9was positively correlated with the AI (r=0.77,P<0.01).ConclusionsInhibitting cell apoptosis may play an important role in the occurrence ofgastric carcinoma. Livin and Caspase-9were closely related to the occurrenceand development of GC. The possible mechanism was influence cellularapoptosis to participate in the occurrence and development of GC.
