High performance capillary electrophoresis (HPCE), founded in the 1980's, is a kind of separating and analytical technique with high performance. Because the mobile phase is electrically driven, there isn't the phenomenon of band broadening in HPCE as in HPLC, and the resolution is higher. At the same time, a great many options make it have a better separation efficiency. In addition, many herits such as low- consumption, rapidness, simpliness, capability of anti-interference and anti-pollution are beyond comparison by liquid chromatography. Owing to these,it has been widely used in the fields such as food,drug, environment, and especially the biochemistry. The following reports were mainly presented for the application and elementary study of HPCE on the pharmaceutical analysis.Part 1. Determination of physico-chemical constants of drugs by high performance capillary electrophoresis.Firstly, capillary zone electrophoresis was used to determine the acid-base dissociation constants of benzoic acid by the method two-point-pH-ratio elimination with salicylate as internal standard. The results indicated that the above method hasn't the drawback that broad range of pH is needed in non-linear regression, which made it more simply, quickly, efficiently, sensitively and accurately to determine the pKa of weak mono-acid. Secondly, logP of several steroids were determined by micellar electrokinetic chromatography(MEKC) so that the hydrophobicity of drugs could be evaluated accurately. All the three different micellar buffer systems showed high similarity to octanol-water system. Compared with the traditional shake-flask method, MEKC is not only simple but also accurate relatively. These experiments proved that capillary electrophoresis is both feasibleand reliable in determinating the physico-chemical constants of drugs .Part 2. Separation and analysis of vitamines by micro-emulsion electrokinetic chromatography(MEEKC).In this part, SC-microemulsion system was firstly used to separate four vitamines. In order to find a new separation mode for multi-component system, the MEEKC method was established by comparing four buffer systems. Because the microemulsion has better resolvent ability and less compatibility, MEEKC showed higher efficiency than MEKC in separating vitamines which have great differences in hydrophobicity. The results suggested SC-microemulsion system has better selectivity than SDS-microemulsion system owing to its benefit for vitamines solved in water.Part 3 Study on fingerprint chromatography and standardization of preparations of drugs.Fingerprint chromatography of Chinese traditional patent drug Jiu Ru Tian Bao Ye was established for the first time. Through identifications of common peaks and typical peaks in Jiu Ru Tian Bao Ye which contained different extracts of four moths respectively, the possibility to identify Jiu Ru Tian Bao Ye by its CZE fingerprint chromatograms was investigated. The results showed the different CZE fingerprint chromatograms of Jiu Ru Tian Bao Ye had distinct characteristics, which indicated CZE fingerprint chromatograms of Jiu Ru Tian Bao Ye could be used to identify this drug.In addition, the standardization method for two preparations of steriods drugs were established by capillary electrophoresis. The results suggested CE not only had good separation power and resolution but also was simple and fast, which could be used to establish standardization method for steroids drugs.
