This paper summarized the investigative status of purifying leukemia cells in vitro by photodynamic therapy(PDT). Considering the importance of light dose in PDT, the research on light dosimetry during laser photodynamic therapy(LPDT) of leukemia was developed preliminarily in theory and experiment.Firstly, the status of PDT was introduce briefly. The light dosimetry during PDT was expound. The difference between the light dosimetry and the distribution of light in biotissue was point out. Monte Carlo simulation can show the distribution of light in biotissue.Secondly, Monte Carlo method was explain detailedly and the distribution of light in marrow was obtained by Monte Carlo simulation. The penetration depth, Ze, was obtained by analyzing the distribution of light in marrow. At the same time, the penetration depth, 6, was calculated by light dosimetry theory. Based on Ze and 6, the backscatter term, k, was gained.Finally, the photodynamic effect experiment with ZnPcS2P2 of K562 cell lines and mononuclear cells was developed. The optimum incubation time, light dose and concentration of photosensitizer were presented. The preliminary result was obtain experiment of leukemia cells in vivo...
