Leukemia is a malignant diseases in hematopoietic system that will doseriously harm to human health. Using photodynamic therapy to purify tumor cellsin vitro is a new method for treatment of leukemia. It uses specific wavelength of lightto irradiate a certain photosensitive material, and then produces a series of chemistry,physics, biological reactions to achieves therapeutic purposes of malignant orbenign diseases.The first part of the disertation narrated the significance of thetopic and the main content. The second part of the disertaton provided an overview ofPDT mechanisms, the factors that affect PDT, and the progress in study of killingtumor cells by TiO2 photocatalytic oxidation. The third part introduced the design andmanufacture of PDT reaction chamber based on LED light source. The fourth partdescribed the principle and operation steps of CCK-8 assays, and experimentallystudied the sensitivity and reliability of CCK-8 in cell activity detection. The fifth partintroduced the preparation of TiO2 and iron-doped TiO2 nanoparticles and its'optical properties. The sixth part researched the effect of TiO2 and iron-dopedTiO2 particles to HL60 cells activity by CCK-8 assay. In the last part, the wholedissertation was summed up.The major achievements made in this dissertation were as follows:1. Designed the photodynamic reaction chamber and set up the entirePDT experimental system. The reaction chamber uses LED as light source.It is stable and juststable in output power. Radiation wavelength can be changed bythe replacement of different LED.2. Participated in the preparation of TiO2 and iron-doped TiO2 nanoparticles, andstudied the character of nanoparticles.3. Did some experimental research on the effect of TiO2 and iron-dopedTiO2 nanpparticles to the HL60 cells activity under different condition, such as incubation time, concentration of nanoparticles, magnetic field strength, light, etc.Based on the experimental results, the mechanism of cells killing by TiO2 andiron-doped TiO2 nanoparticles was discussed.
