| ObjectiveTo investigate the effect of the Heweichongji (HWCJ, composing of amomum, chicken gizzard membrane , medicated leaven and malt) on the gastrointestinal motility of normal and dysmotility rats, to select the best among HWCJs containing three different amonum respectively , and to study its influnces on the plasma motilin and nitric oxide of rats as well as its acute toxicity.MethodsAll experimental animals were fed for one week. The gastric emptying and small intestinal propulsion were observed by the method of nutritional semi-solid feeding. Dysmotility models were induced by intraperitoneal injection of L-argine or L-ephedrine . Radioimmunoas-say and Griess method were used to determine motilin and nitric oxide in experimental animal plasma respectively. The toxicity was evaluated by meams of max endured multiple.ResultsCompared with negative control, the gastrointestinal motility of all kinds of HWCJs was significantly improved (P<0.05). There was no statistical difference between HWCJ and Cisapride (P>0.05) as well as among different kinds of HWCJs(P>0.05),however the drug with amomum villosum lour was better than others. The gastrointestinal motility did not significantly increase as the dose increasd . HWCJ was able to antagonize the hypomotility caused by L-argine ,but had no influence on the model induced by L-ephedrine. HWCJ markedly increased the plasma motilin . The values were significantly greater than negative control (P<0.01). It had no effect on nitric oxide of experimental animals(P>0.05 ). All of rats in the toxicity experiment survived and the max endured multiple was 240.ConclutionHeweichongji can improve gastroduodenal motility effectively. The HWCJ with Amomun villosum Lour is more effective than others, but dose-response relationship is not noted. The mechanism of improving gastrointestinal motility of HWCJ is not due to the adrenal receptor , but plasma motilin increased and nitric oxide regulated. HWCJ has little toxicity. |
