PrefaceFunctional Dyspepsia(FD)is a common syndrome of digestive,which pathogenesis has not completely unawared.Nowadays,treatments on this disease of West medcine is just drugs that could simply stimulate gastrointestinal Motility,for example, Domp-eridone and Itopride.As National "the Eleventh-Five-Year Plan"Spleen-and-Stomach disease specialist,our department has made success on cure of FD with Traditional Chinese Medicine.After many clinical researches,we concluded that the syndrome of "Spleen Qi Stagnation"is the main TCM syndrome.Weitongxiaopijiajian Decoction is a decoction that plus and minus some herbs on the foundation of the classical Decoction of Sijunzi decoction,which can stimulate gastrointestinal Motility,and can release the stagnation of Qi to help digest food as well.This experiment aims at discovering the way that WTXPJJ decoction affect experimental animals'gastrointestinal Motility and hormone.ObjectiveTo observe the affection of WTXPJJ Decoction on plasm MTLilin(MTL), Gastrin (GAS),serum Nitric Oxide(NO),Gastric emptying rate and Intestinal propulsive ratio in functional dyspepsia(FD) rats,on the expectance of discovering the way that WTXPJJF affect experimental animals'gastrointestinal Motility and hormone,and the relationship between the affection and different TCM dose.MethodsSixty healthy male wista rats were randomly divided into five groups:Control group,Model group, WTXPJJ low dose(6.25g/kg,middle dose(12.5g/kg),large dose group(18.75g/kg) and Domperidone group.Different drug were given to six groups: Normal sodium,Different concentrations of WTXPJJ Decoction and Domperidone distilled water solution once a day by gastrogavage for 14days; Then the plasm MTL, GAS, serum NO,and the rate of gastric emptying and Intestinal propulsive ratio were observed.Let the rats live freedom during the first week to make sure that the rats adjust to the new environment.In the next ten days,the rats were gastric injected into 4℃vinegar to establish the FD model.Then corresponding drugs were given to those five groups: Normal sodium,Different concentrations of WTXPJJ Decoction and Domperidone distilled water solution once a day by gastrogavage for 14days.18 hours before the last time to give drug,food was forbidden for those rats,but not water.Then 2 hours after the last time drug,we took blood of those rats,and separated the plasma and serum.To calculate the content of MTL and GAS on the methods of radioimmunoassay according to the instructionThe content of NO was exzamined by the methods of Nitrate reductase.Rats in each group were gastric injected the nutritional semi-solid 0.3mL/100g gavage,which was mixed with ink,according to rat's body weight.30mins later,rats were killed Cervical-dislocatly, and then we calculated the rate of gastric emptying and intestinal propulsive ratio.Results1.Affections on Plasma MTLinlinThere are statistical significance between each treatment group and the model group on MTL level(P<0.01).The MTL level of both middle and large dose group are significant higher than the Domperidone group(P<0. 01),but the difference between the two group has no significance(P>0.05). Compared with the Domperidone group,the low dose has no significance(P>0.05).The control group's MTLinlin is higher than the modle group, but lower than the middle and large dose group(P<0.01);while compared to the Domperidone and the small dose group, the differences have no significance(P>0.05).2.Affections on Plasma GastrinEvery treatment groups'GAS level are significantly higher than the model group(P<0.01).Every dose of WTXPJJ decoction's GAS level are higher than that of the Domperidone group(P<0.01).The middle group's GAS level is higher than the other WTXPJJ decoction group(P<0.01). The control group's GAS is higher than the modle group, but lower than the small,lmiddle and large dose group(P<0.01, P<0.05);while compared to the Domperidone group, the difference has no significance(P>0.05).3.Affections on Serum Nitric OxideEach treatment groups'NO level are significantly lower than the model group(P<0.01).Both the middle and high dose groups' NO level are lower than that of the Domperidone group(P<0.05),but no significance between the middle and high dose group(P>0.05).Compared with the Domperidone group,the low dose group's NO has no significance(P>0.05). The control group's NO is lower than the modle group(P<0.01),but have no significances when compared to the other groups(P>0.05).4.Affections on the Rate of Gastric EmptyingEach treatment groups'GER are significantly higher than the model group(P<0.01). Compared with the Domperidone group,the GER of every WTXPJJ decoction there are no significance(P>0.05). The control group's GER is higher than the modle group, but lower than every treatment groups(P<0.01).5.Affections on the Intestinal Propulsive RatioEach treatment groups' IPR are significantly higher than the model group(P<0.01).Both the middle and high dose groups' IPR are higher than that of the Domperidone group(P<0.01),but no significance between the middle and high dose group(P>0.05). While the low dose group's IPR is lower than that of the Domperidone group(P<0.05).The control group's IPR is higher than the modle group (P<0.01);while compared to the middlle and high dose group, the differences have no significance(P<0.05).Conclusions1.Every dose group has the function of increasing the MTL,GAS,IPR and decreasing NO,which is better than that of the Modle group(P<0.05orP<0.01). 2.Every dose group can increase the GER(P<0.05).3.The function of stimulate gastrointestinal Motility and secretion hormone of the middle group is better than that of the low and high dose group.
