Through text of Fas/Fasl in the patient with different pathology grade and text of the relation between the apoptosis and the extent of malignant tumor, the effect of Fas/Fasl in the growing of gastrocytoma cell tumour and development have been analysed.MethodThe expression intensity of the Fas and FasL in different pathology grade of astrocytoma has been texted by immunohistoehemistry ( S - P method ). The experiment sample were collected from the second clinical college neurosurgery in the China medical university on Febu-rary, from 1999 to 2002 January, totally 50 examples, aging from 6 years to 73 years, 29 male and 21 female, among them, including41 astrocytoma examples. According to the tumour classifying standard of neuroepithellal tissue by WHO, which is divided into; grade I 10 case, grade II 10 case, grade III 11 case and grad IV 10 case. All patients are not cured by chemotherapy, telelherapy and immunity before operation. All specimens are confirmed astrocytoma through pathology.The specimen is all fixed 48 hours through 10% formalin, andthe paraffin wax embedding. The 4jxm of every example paraffin wax section continuously has been cut into slices, and is used immunohis-tochemistry dye after antigen heat repair. The normal brain tissue is used as the contrast group. The positive contrast of Fas and FasL is lymphoma chosen through pathology diagnosis. The positive cell of Fas and FasL is brown - yellow particles scattered within cytoplasm or cell membrane, 200 cells are observed under 400 times microscopes, the negative cell don not have brown - yellow particles. The positive cell number occupies total tumor cell <5% ,5 -25% , > 26% , serve as ( + ) , ( ++ ) and ( +++ ) respectively.The statistics processing; The expression of Fas/FasL is adopted X N related analysis. The above analysis is progressed with statistics software SPSS.ResultsThe positive matter of Fas/FasL by immunohistochemistry assumes brown - yellow particles, and its expression is negative in normal brain tissues. Positive matter of Fas and FasL is in cytoplasm or cell membrane of astrocytoma. The expressing rate of positive cell is 68. 3 % ( 28/41) and 75. 6 % ( 31 /41) respectively in 41 case astrocytoma. There are 28 and 31 different extent expression of Fas/FasL in 41 case astrocytoma, and the expression of Fas/FasL in astrocytoma has certainly relation with their pathology grade. With the malignant extent of astrocytoma gets higher, the expression of Fas/FasL also goes up. In 41 case, the expression of Fas/fasL which is - or + in 1 ~ 2 grades astrocytoma occupies 95. 0% and 75.0% respectively, and occupies 71.4% and 66. 7% respectively in 3 ~4 grades of astrocyto-ma. The statistic analysis shows that the expression intensity of Fas/ FasL is mutually related with pathology grades of astrocytoma. The positive area of Fas/FasL expression concentrates around ischemia and necrosis area after the pathology section in 41 example patients, and occupies 92.9% and 96. 8% respectively. The proportion of ischemia and necrosis increase correspondingly with the malignant extent of astrocytoma. The observation of radio in the set of patient ( CT or MRI ) shows that 13 case have strengthen effect before the operation of tumor, among them the positive case of Fas / FasL is 46.2% and 61. 5% respectively. There isn't significantly different by analyzing statistics.DiscussionThe apoptosis is one kind of cell death distinguishing from cell necrosis, under fixed physiology and the pathology condition, it is one kind of programmed cell death ( PCD) , and it does not cause organism inflammation. The Fas is an important factor to regulate and control cell apoptosis. The Fas unusual expression has the very close relation with the tumor occurring and developing.Fas express in many tissues, including chest gland, liver, cardiac muscle, skin as well as ovary, but does not express at the central nervous system.
