Experimental Study Of Antitumor Effect Of Chinese Herbal Prescription "Niu Huang Tian Long Yin" On Apoptosis Of SK Tumor Cell And S-180 Tumor Cell

Objective: In this study, the toxicity and antitumor effect of Chinese herbal prescription "Niu Huang Tian Long Yin" were investigated. The prescription is based on the troditional chinese herbal prescription "Xi Huang Wan", combined with Hirudo, Fructus crotonis and so on.their effects were observed on the mice transplanted with the tumor cell line of S-180 and SK.Methords: A small amout mice were given the chinese herbal prescription pre os, and observed the presence after be poisoned, the 0% and 100% estimated half lethal dose (mini and maxi) were found out. So did a large number mice the half lethal dose (LD50)was worked out. The cold storage tumor cell line (SK and S-180) were reanimated fast and subcultured. When the tumor cell were cultured to a large amount, then transplanted into the mice which made the mice grow out the tumor tissue. The mice transplanted with the tumor cell were given the chinses herbal prescriptio, and were put to death after 12 days, the tumor tissues were get out and compared the tumor weight with each group. The tumor tissues' morphylogical change were observed by electron micrescope. The pattern andkinetics of cell cycle of these tumor cells were analysed by flow cytometry.Result: From the acute toxicity test, worked out the scope of 0% and 100% estimate half lethal dose is 40g/kg to 90g/kg, and the half lethal dose(LD50) is 58.5g/kg. According to the acute toxicity test identify standard, the chinese herbal prescription is non-toxic pre os.The results showd that the prescription coulde induce apoptosis of tumor cells in vivo, inhibit the growth of tumor tissue of mice transpanted with SK and S-180 cell line. And the growth inhibit rates to SK were 78.27%, 71.36%, 41.77%, respectivity (P<0.01); and that to Sk were 46.09%, 44.95%, 37.12% respectivity (P<0.05); The morphylogical change were observed by means of electron microscope. The typical characteristics of apoptosis were found, for example, cell volume reduction, condensation of cytoplasm and chromatin, and fragmentation of DNA at internucleosomal sites, plasma membrane blebbing and apoptotic bodies formation etc. The pattern and kinetics of cell cycle of these tumor cells were analysed by flow cytometry, and typical apoptotic peaks were noted, and apoptosis rates of tumor cells in experimental groups were increased considerably(P<0.05) and synthesis of DNA of tumor cells were inhibited obviously, the cell numbers in the S and G2M phase were increased, so the proliferation index(PI) were reduced. All the results showed that the Chinense herbalprescription "Niu Huang Tian Long Yin" had antitumor effects on various tumor cell lines (S-180 and SK), which provided theory basis for using chemotherapeutic drugs together with cytokines reasonably and using "Niu Huang Tian Long Yin" to treat patients with cancers in clinical practice, especially the antitumor effects of "Niu Huang Tian Long Yin" deserve to be studied, developed and applied in the future.
