Objective: The tonic and beneficial traditional Chinese medicine has the effect of improving human immunity and anti-cancer by promoting proliferation of immunocytes and cytokines. But it is still in question if it is of benefit to the growth of cancer cells through increasing immunity. The objective of this study is to observe the effect of tonic and beneficial traditional Chinese medicine on the growth of human esophageal carcinoma cells and on cancer tissue by experiment in vivo and vitro and flow cytometry. The study is to give some advices for clinical use.Materials and Methods: 1. Cell cultivation in vivo: The tonic and beneficial Chinese medicine of different concentration ( Huangqi solution and Shenqi solution) was put into culture medium with the TE-13 cell line of human esophageal carcinoma which was in the logarithmic growth period. Then the cells were cultivated in the CO2 culture box. On the third day the cultivation liquid and the Chinese medicine of corresponding concentration were changed. And on the sixth day, after digestion, centrifugation and staining by placental blue, the cells were counted under the optical microscope in order to observe and compare the effect of thetonic and beneficial Chinese medicine of different concentration on the TE-13 cell line of human esophageal carcinoma. 2. Animal experiment: T739 mice with weight from 18 to 22 grams (Half in female and half in male) were used. Ten mice were included in each group. The mice models either with smaller size or larger size were prepared with LA795 lung adenocarcinoma cell strain of mice (from the animal center, Tumor Hospital, Chinese Academy of Medical Science). The mice with LA795 lung adenocarcinoma cell were put into death. Then fresh tumor tissue was obtained and prepared into the single cell liquid of 3×107/ml. 0.1ml of the liquid with cells of 3×106 was injected between the toes of left lower limb of mice. And the mice model with different load of tumor was prepared. In Group one, as soon as the tumor nodule was larger enough to see, the Chinese medicine was injected intraperitoneally. And in Group two, when the average geometric diameter of tumor was 5.0±0.2ml, the Chinese medicine was used. In the control group 0.2 ml of saline was injected intraperitoneally ,and 0.2 ml of Huangqi solution and 0.2 ml of Shenqi solution were injected intraperitoneally respectively in the study group. Thirty days were needed in each group. Every day the mice weight was measured by electronic balance and the tumor volume was estimated by electronic calipers in order to observe the differences of the weight of mice, tumor volume and survival time between thecontrol group and the study group when the mice were in the different load of tumor. In addition, the effect of the tonic and beneficial Chinese medicine the weight of mice, tumor diameter, survival time when combined with radiotherapy were observed using the same method mentioned before. 3. Flow cytometry: The mice with tumor were put to death after 7 days' intraperitoneal injection of the tonic and beneficial Chinese medicine. Marginal tissue of tumor was obtained, fixed by 70% alcohol and estimated by FCM in order to observe the apoptotic index, cell proliferation index and VEGF after application of the tonic and beneficial Chinese medicine.Results: 1. Cell cultivation in vitro: 1.1 The cell count of TE-13 cell line of human esophageal carcinoma was done after 6 days' application of Huangqi solution of different concentration. The cell growth of Study Group 2 and Study Group 3(The concentration of Huangqi was 3.45% and 1.7% respectively) was much faster than that of the control group, with statistical significance of P < 0.05. But there was no significant difference between the cell growth of Study Group 1 with Huangqi solution of 6.9% and the control group. 1.2 The cell count of TE-13 cell line of human esophageal carcinoma was done after 6 days' application of Shenqi solution of different concentration.
