Objective To detect the cell cycle of nasal tumor by FCM,and explore the significance of DNA index(DI) and S-phase fraction(SPF) in the diagnosis of nasal lesions.Method From all who underwent surgery in otorhinolaryngology department of Qingdao Municipal Hospital during 2002 to 2006, 30 sino-nasal malignant tumor,30 nasal inverted papilloma were chosen as experimental group, and another 30 cases of normal mucosa as control group. The paraffin- embeded specimens were cut into slices of 50 micrometers,and prepared a single cell suspension. The cell cycle was detected by flow cytometry. The DNA index,S-phase fraction and heteroploid rate were analyzed by statistical treatment. The variation of heteroploid rate in different tissues and the correlations of DNA index,S-phase fraction with tumor characteristics were investigated.Results DNA index represents the DNA content and S-phase fraction represents the cell proliferation index .The DNA index and S-phase fraction of the control group was 0.99±0.16, 9.94±2.57, the nasal inverted papilloma group was 1.03±0.15, 10.67±2.63, sino-nasal malignant tumor was 1.03±0.15, 10.67±2.63 respectively .Compared the nasal inverted papilloma group with the control group, the DI was higher but without statistical significance , and the SPF was statistically higher than that in the control group. The DI and SPF of malignant tumor were significantly higher than that of the control group and that of the nasal inverted papilloma group.DI= 1.0±0.1 was judged on distinguishing diploid from heteroploid .There was no heteroploid in the control group,4 heteroploid in the nasal inverted papilloma group , and 21 heteroploid in the malignant tumor group. Statistics analysis showed: comparing the nasal inverted papilloma with the control group, P>0.05, the differenc of heteroploid rate were with no statistical significance; compareing the malignant tumor group with the control group and the nasal inverted papilloma group respectively, the heteroploid rate were statistically different. While, the S-phase fraction of the heteroploid group was statistically higher than that of diploid group.Conclusion Flow cytometric analysis of DNA index,S-phase fraction could be a reference index of judging the nasal tumor characteristics, and also be reference valuable to conjecture the evolution of inverted papilloma.
