Objective The telomerase activity of peripheral blood lymphocytes was measured by polymerase chain reaction-based enzyme immunoassay(PCR-ELISA) assay. To investigate the relation between telomerase activation, abnormal telomere length and blood vessel cell of patients with essential hypertension senescence , abnormal proliferation; and the effect of antihypertensive treatment on telomerase activity. To provide a theoretical basis for study the pathogenesis in essential hypertension.Methods After medical history investigation and physical check-up, the test group was drawn blood of five milliliters by heparin resisting concrete. By normal centrifugal action peripheral blood lymphocytes were broken up with lymphocytes separation medium. The telomerase activity of lymphocytes was assayed by polymerase chain reaction-based enzyme immunoassay (PCR-ELISA).Results The telomerase activity of peripheral blood lymphocytes in hypertensive group was singnificantly increased than that in control group(0.45±0.22 and 0.24±0.11,respectively, P<0.001). The increased activity of telomerase were found in first stage and low-risk hypentersives. According to level of hypertension ,telomerase activity of peripheral blood lymphocytes was upregulated to the same level in every group(P>0.05). According to prognostic risk factors, telomerase activity of peripheral blood lymphocytes was upregulated tothe same level in every group(P>0.05). Telomerase activity of peripheral blood lymphocytes was upregulated to the same level in the group of regular antihypertensive treatment and the control group(P>0.05).Conclusion Telomerase activity was detectable at low levels in peripheral blood lymphocytes of healthy subjects. The telomerase activity of peripheral blood lymphocytes in hypertensive group was increased. It was found that the increased telomerase activity were not correlated with the severity of hypertension and prognostic risk factors. The telomerase activity was decreased after antihypertensive treatment.
