Sappan Lignum is dried heartwood of Caesalpinia sappan L.. It has been used in oriental medicine as an analgesic and anti-inflammatory agent for a long time in China. It has high bioactivities and wide utilization. Now it is found Sappan Lignum has immunosuppressive function in acute rejection after allograft heart transplantation.This paper deals with the immunosuppressive function of compounds obtained from the extract of Sappan Lignum. It has been extracted, isolated and identified on the base of the determination of bioactivities by the method of pharmacodynamics. In the course of screening for immunosuppressive function of extract from the plant by several rat allograft heterotopic heart transplantation models and mixed lymphocyte culture experiments, the alcoholic extract is found that it has a significant activity. By column chromatography with silica-gel and recersed phase-gel, nine compounds were isolated finally. On the basis of chemical and spectral analysis ,the structure of 3 compounds were identified as: brazilin(compound 1) % protosappanin A (compound 2)and 7-hydroxy-3- (3' ,4' -dihydroxybenzylidene)-chroman-4-one(compound 3).The compound 1 and 3 were isolated from Sappan Lignun for the first time in China. The result provides important scientific foundation and material base for further research on bioactivities, pharmacological action theory, preparation and development of new drug for immunosuppressive function in acute rejection after heart transplantation.
