PREFACEAcute pancreatitis is a common but rather dangerous clinical disease, usually with MOSF and severe systemic infection at early stage and leading to high death rate. The mechanism of acute pancreatitis is extremely complicated. It was once considered that pancreas activation was the original factor of acute pancreatitis, but no ideal effect could be attained through using inhibitor of pancreatic secretion and protelyt-ic enzymes. However, the latest research shows that inflammatory mediators play important role in the mechanism of acute pancreatitis and in the evolutionary process from acute pancreatitis to acute necrotizing pancreatitis.ICAM-1 is a biologically active substance with great importance found recent years. The research on its involvement in systemic inflammatory reaction has become the focus. It is playing an important role in many aspects, such as maintaining the stabilization of normal tissues, involving in the transfer of inflammatory cells, forming thrombus and transferring tumor. It calls the attentions of many schol-ars all over the world.Many experiments show that the movement of PMN is the fundamental proof of the incidence acute pancreatitis while the precisely reactionary mechanism is yet unknown. The inflammation which is caused by the accumulation and infiltration of PMN partly depends on the expression of P-selectin and ICAM-1. The immune neutralizing function has been proved to be protective during a series of laboratory acute pancreatitis. Owing to P-selectin's important role in the incidence mechanism of acute pancreatitis. The study on the role of PPS in the diagnosis of acute pancreatitis, the judgment of the severity of acute pancreatitis and the prediction from acute pancreatitis to acute necrotizing pancreatitis, has been further developed and gradually calling public attention. However, during the experimental process from acute pancreatitis to acute necrotizing pancreatitis on rats, the variation of the expression of P-selectin in pancreas, the alteration of the concentration of PPS and the expression of P-selectin after the use of monoclonal antibodies that all these serial research are seldom reported.OBJECTIVESThrough the establishment of acute pancreatitis rat model, during various periods of time inducing acute pancreatitis, we are to observe the dynamic change of P-selectin in pancreas and PPS. Also after the monoclonal antibodies injections into rats, we are to observe the change of P-selectin, PPS mentioned above. Then we discuss the relationship between the expression of P-selectin and acute pancreatitis, and thus provide a new target for acute pancreatitis diagnosis in clini-cal work, prognosis judgment and effective cure.MATERIALS and METHODSExperimental Materials; 1. chief experiment materials and drugs: Sprague-Dawley rat; Poly-L-lysine; GMP-140 kit; anti-P-selectin monoclonal antibodies; SP immunohistochemical kit; 5% sodium tau-rocholate; 2% EDTA; Olympus microscope. Methods: SD rats were anesthetized with an intraperitoneal injection of 10% urethane. pancreatitis was induced by retrograde injection into the biliopancreatic duct of 5% sodium taurocholate in a volume of 0. 1 rnl/100g bodywet using an infusion pump. Control animals received an intraductal infusion of saline solution. 1,3 and 6 hours after induction, plasma,pancreas tissue samples were obtained. The plasma samples were obtained to analyze amylase and PPS. The pancreas tissue samples were obtained to examine the expression of P-selectin.RESULTSWhen injected in sodium taurocholate, the pancreas thus became swollen, followed by hemorrhage and putrescence sporatically saponif-teated spots in peri-pancrease and mesentery, gradually aggravated as-cites after the incidence of acute pancreatitis for 3 hours. When the model was induced for one hour, swollen pancreas cells could be seen through microscopes, and the leukocytes could be seen permeated and spotted putrescence in pancreas; when the model was induced for 3 hours, extremely swollen pancreas cells could be...
