Objective : Diabetes mellitus is a common endocrine disease due to metabolic disordor. It's chronic complications invade the tissues of cardiovascular, kidney ,nerves, retina ect,and especially can cause bone disorder, which at last leads to osteoporosis .Recently, many studies show that the incidence of diabetic osteoporosis(DO) was much higher than other chronic complications of diabetes. DO will cause pain in many bones of the body which is likely to cause fracture . Accordingly ,it has high incidence of disability and high mortality as well .Now close attention has been paid for DO by many people. Although the present research works on DO have already made progress to some extent ,its pathological mechanism is still a mystery and it cann't be cured by some ideal treatments due to its complicated promoting factors and routes. So ,it has been an important task to elucidate DO's pathological mechanism in order to develop efficient medicines for clinical treatment. In recent years ,a lot of research work has been done from basic research to clinical research about the pathological mechanism of diabetic chronic complications. The realization to the mechanism is more and moredistinct. Synthesizing these research results , a kind of substances, which are called advanced glycation end products (AGE),have been found to be the common substances for all diabetic chronic complications .The effect of AGE on cardiovascular disorders and kidney disorders has been recognized. There have been many studies about how to prevent and treat diabetic chronic complications by getting rid of the overproducts of AGE and inhibiting the formation of AGE, some of which have gone into second clinical research . However, the research about the effect and mechanism of AGE on DO has still not been reported . Our research team has got some delectable research results . It is the first that Dr. Dejuan Kong found that between the amount of AGE in bone and the BMD existed active linear dependence.When we used aminoguanidine (AG),the amount of AGE in bone decreased and the BMD sharply increased ,which showed that by inhibiting the formation of AGE could prevent and treat DO. Although AG has the better effect, it has been said that high concentration of AG may lower the sensitivity of pancreas tissues to hyperglycemia and inhibit the production of insulin. At present, AG hasn't been reported to be used in clinic. For this ,utilizing this mechanism of AGE, we can develop effective herbs,which will exert the superiority of Traditional Chinese Medicine (TCM) and provide a new means for preventing and treating DO. In this study ,we established the model of diabetic rats,and prepared the polyclonal anti-AGE antibody, and examine the BDM of diabetic rats and the amount of type I collagen and AGE in their bones , in which therole of AGE on DO has been discussed fartherly. Furthermore, compared with AG, tonifying kidney herbs's inhibiting effect on the formation of AGE was studied for preventing and treating DO.Method: From forty-eight female Sprague-Dawley rats (six months of age ),twelve rats were chosen at random as normal control group (group CG).The rest were given an i.p.injection of strepozotocin(STZ)(40mg/kg).All rats with plasma glucose level≥13.8mmol/L after 72 hours were included in the study.These rats were divided into three groups at random:diabetic group(group CG), aminoguandine-treated diabetic rats(group AG );herb-treated diabetic rats(group HD);diabetic rats group (group DM).The rats of group AG, group HD and group DM were injected with STZ (40mg/kg avoirdupois, being dissolved into buffer , pH4.4, 10mg/ml concentration) to establish the model of diabetes. All were provided normal feed .Rats of group CG and group DM drank water freely . According to body surface area converting formulae between human and rat ,the rats of group HD were filled stomach with herbs(1g/ml).A rat was filled 3ml everyday.
