OBJECTIVE AND BACKGROUNDDiabetic cardiovascular disease is one of the most severe diabetic complications, which is responsible for many of the disabilities and short life expectancy associated with diabetes mellitus.As is diabetes itself,diabetic cardiovascular disease is a multifactor disease.Studies have revealed that the advanced glycosylation end products (AGEs)participate in the pathogenesis of diabetic cardiovascular disease through several mechanisms,including their binding the specific receptors(RAGE).Therefore, limiting RAGE expression might be an intriguing concept to modulate cardiovascular disease in diabetic patients.The protective effect of Puerarin,an isoflavone purified from Chinese herb radix of Pueraria lobata,is widely used in cardiovascular disease therapy for its dilatation of the coronary artery and antioxidant effects.Recent studies have revealed that Puerarine can reduce RAGE expression in human endothelial cells,thus limiting the cells' susceptibility toward proinflammatory AGE effects.The aim of the present study was to determine whether Puerarine affects RAGE mRNA in cardiovascular tissue of streptozotocin-induced diabetic rats.RESERCH DESIGN AND METHODSWe induced Diabetic rats by an intraperitoneal injection of STZ in SD rats.The criterion of diagnosis of diabetes was the consecutive blood glucose level≥16.7 retool/L.30 male SD rats were randomly divided into 3 groups,Diabetes adding Puerarine group(n=11,intraperitoneal injection Puerarine 100mg/kg.d)and Diabetes adding BBS group(n=11)and normal control group(n=8).The body weight and blood glucose were measured every two weeks.8 weeks later,all rats were killed and measured serum creatine kinase-MB(CK-MB)levels.The expression of RAGE mRNA was quantified in cardiovascular tissues by reverse transcription-polymerase chain reaction(RT-PCR)respectively.RESULTS1.The RAGE mRNA expression was increased in cardiovascular tissue of diabetic rats.RAGE/β-aetin ratio of diabetic group was significantly higher than that of the control(P<0.01).2.After treatment with Puerarine,The serum CK-MB levels were lower than diabetic group.Also,RAGE mRNA expression decreased significantly and RAGE/β-actin ratio was obviously lower than that of the control.3.After treatment with Puerarine,the blood glucose was lower than that of diabetic rats.CONCLUSIONS1.The high expression of RAGE mRNA may participate in the cardiovascular damage of diabetic rats.2.Puerarine can protect the cardiovascular tissue from the impairment of hyperglycemia and advanced glycation end product.3.Puerarine can decline the blood glucose of diabetic rats.
