The oral cavity and maxillofacial region are the exposure region of the body, which is often injuried in accident conditions, such as trauma, traffic accident, the war and the quality of wound healing directly impacts the beauty of patiens' faces. In oral cavity and maxillofacial region there are rich sensory nerves. The neuropeptide(SP) secreted from sensory nervous terminals affects the quality and the speed of wound healing. The substance P(SP)mostly consists in the terminals of two kinds fibers, the thinly myelinated Aδpain fibers and the unmyelinated C fibers. During the course of wound healing, SP takes part in the proliferation and modulation of the repairing cells.Our previous experiments in vivo and in vitro showed that substance P could facilitate wound healing, which was related with the effect of substance P on the expressions of endogenous growth factors and their receptors in tissue fibroblasts. But the signal pathway by which the substance P regulates the expressions of endogenous growth factors is not clear yet. Recently some of investigations demonstrate: some of oncogenes not only are activated during the course of cell proliferation, but also influence cellular proliferation and differentiation at the levels of ligands interacting with receptors, biochemistry alteration resulting from the signal transduction and gene transcription inside cells. In a ward, SP is very close correlation with tissue repair. As the main component cells of granulation tissue--the fibroblast plays the important role in processes and outcome of trauma repairing.Research of DNA sequence showed that bFGF,TGF genes contains the combined sites for TRE. These sites could be recognized and combined by AP-1 which is a heterogeneous dimmer is constituted by c-fos and c-jun and is able to activate transcription of corresponding genes. To investigate the roles of signal transduction of c-fos and c-jun on regulation of substance P for expressions of endogenous growth factors, a cultured fibroblast model in vitro of rat granulation tissue was established in order to eliminate interferes of multiple factors in vivo. The techniques of immunohistochemistry and hybridization in situ were used to detect mRNA and protein expressions of c-fos and c-jun. Our study may provid theoretic basis to improve nerve's function, to regulate secretion of neuropeptides and to facilitate wound healing.The Investigating results showing:(1)The c-fos mRNA and protein expression offibroblasts markedly displays time-effect relationship with the stimulation of the SP. The c-fos mRNA expression of fibroblasts in experiment group increased obviously from 25 minuts to 1 hour after stimulating cells with the SP 10-7M; The c-fos protein expression increased markly at 2 hours after stimulation with the SP, and showing a peak at 2 hours, persisting in 3 hours, after 5 hours, there is no discrepancy between the experiment group with the control group.(2)The c-jun mRNA and protein expression of fibroblasts markedly displays time-effect relationship with the stimulation of the SP. The c-jun mRNA expression of fibroblasts in experiment group up-regulated obviously from 25 minuts to 2 hour after stimulating cells with the SP 10-7M. The level of the c-jun mRNA decreased to normal at 3 hours; The c-jun protein expression increased significently from 2 hours to 3 hours after stimulation with the SP, decreasing to the normal level at 5 hours.(3)The bFGF mRNA and protein expression of fibroblasts markedly displays time-effect relationship with the stimulation of the SP. Compared with the control group, the bFGF mRNA expression of fibroblasts in experiment group up-regulated significently from 5 hoursto 8 hour after stimulating cells with the SP 10-7M. The level of the bFGF mRNA decreased to normal at 10 hours; The bFGF protein expression increased markly from 12 hours to 18 hours after stimulation with the SP, decreasing to the normal level at 24 hours.(4)The TGFβ-1 mRNA and protein expression of fibroblasts markedly displays time-effect relations...
