Objective: to study the effects of dietary fiber(DF) in Oat bran on blood glucose and blood lipid in rats with experiment type 2 diabetes mellitus and T2DM patients Methods: this study includes two parts. Part I : Animal experiment. 50 wistar rats(194.4 5.3g) are divided into 5 groups base on their body weight. Every group has 10 rats (half male and half female). The control group (group A) is fed normal diet. The other four groups are injected with lower dose (25mg/kg bw) of streptozotocin (STZ) and fed a diet with high calorie and high fat. After formation of the diabetes model, group B (T2DM model group) is fed a diet with high calorie and high fat, and group C, D, E are fed 30% DF, 40% DF and 60% DF respectively. The effects of DF on metabolism of glucose and lipid , and availability of food are observed. Part II: clinical study . 40 diagnosed T2DM patients are chosen and randomly in addition to medicine treatment, the intervention group is given the same treatments except DF. The effects of DF on metabolism of glucose and lipid in diabetes are observed. Results: 1. the difference of food availability is statistically significant. Compared group B with the other four groups respectively , the differences are all significant 2.Before the injection of STZ(OW), the level of blood glucose and lipid in every group are statistically same. But on the eighth week the threeconcentration of DF all decrease fasting blood glucose (FBG), Triglycerides, (TG), Total cholesterol(TC), and Low density lipid-cholesterol (LDL-C), significantly, and increase High density lipid-cholesterol (HDL-c) at the same time. The effects of 40%, 60% DF stronger than 30% DF. 3.the glucose tolerance curve of every group is significantly different. The curve of group A and B are steep, while group C,D and E are relatively smooth. 4. Effects on pancreas tissue: the pancreas of group A rats are normal, and group B are damaged at different level. The majority pancreas of group D and E are normal. The clinical study states that DF can change the FBG, Postprandial blood glucose( PPG), TC, TG, LDL-c and HDL-c as well at the 8?week both in control and intervention groups, moreover, the different of Ow and 8w in intervention groups are all statistically greater than those in control groups, that indicates the intervention can decrease the blood glucose and blood lipid significantly. Conclusion: DF can decrease the blood glucose and blood lipid.
