purpose: To study the relationship between the expression of bcl-2 and mt-P53 protein in tumor tissue, intratumoral microvessl density (MVD), the clinical pathological characteristics and the survival time of patients with lung cancer and observe the relationship within MVD,bcl-2,mt-P53 , meanwhile, to discuss its clinical significance.Methods: The expression of bcl-2 and mt-P53 in cancer tissue were measured by s-p immunohistochemical method in 60 cases of human lung cancer, meanwhile, anti-CD34 monoclonal antibody directed against endothelial marker was used to assay MVD.Results: (1) MVD was significantly associated with lymph node metastasis ( p<0.05 ) , and clinical stage (p<0.05);The prognosis for patients with high MVD was significantly poorer than that of patients with low MVD (x =7.24, p<0.01). Multivariable COX model analysis indicated the MVD were an independent predictor of survival. Significant correlation between bcl-2 expression and histological type of lung cancer (p<0.0 1), differentiation of non-small cell lung cancer (NSCLC) was detected (p<0.05 ) .The expression of bcl-2 protein was not related with lymph node metastasis and clinical stage; In 60 cases ,mt-P53 positive rate in lymph nodemetastasis group (64.29%) was significantly higher than that of the non-metastasis group ( 34.38% ) . No association was noted between the expression of mt-P53 and histological type of lung cancer, differentiation of NSCLC, clinical stage. COX model analysis indicated that no association was noted between the expression of mt-p53, bcl-2 and survival. (2) In non-small -cell lung carcinoma, patients with bcl-2 -negative had significantly higher MVD than those with bcl-2-positive ones ( p<0.05 ) .In addition, there was a significant inverse correlation between bcl-2 and MVD (r= -0.375, p< 0.01 ) ; On the contrary, patients with mt-p52 positive had significantly higher MVD than those with mt-P53 negative ones (p<0.01) .Significant positive correlation was observed between MVD level and mt-P53 level (r= 0.051, p< 0.01) . (3) The mt-P53 positive rate in bcl-2-negative group (77.42%) was higher than that of bcl-2-positive group (26.32%) , bcl-2 and mt-P53 protein expression had correlation (p<0.05) .In bcl-2-negative patients, MVD of cases with mt-P53-positve was significantly higher than that of cases with mt-P53-negative (p<0.05 ) .But in bcl-2-positive patients , the MVD of cases with mt-P53-positve was similar to that of cases with mt-P53-negative.Conclusion: ( 1 ) MVD were the significant variables for predicting prognosis. (2) There was correlation between mt-P53 and lymph node metastasis of lung cancer, bcl-2 detection by s-p is useful in predicting themalignant extent in NSCLC . (3 ) These findings indicated that bcl-2 may inhibit angiogenesis, mt-P53 could down regulate bcl-2 to promote angiogenesis.
