Inhibitory Effect Of Zhibie Capsule (ZBC) On Hepatic Fibrosis In Rats

Objective: To explore the effect and mechanism of Zhibie Capsule (ZBC) on anti-hepatofibrosis.Method: We chose 66 healthy clean rats, the female and male rats are 50 percentage of them respectively. They were divided into normal group, model group, Colchicine group, there different dose ZBC groups. The model of hepatic fibrosis(HF) was induced by complex factors of CCL4, foods of high fat and alcohol. At the end of inducing model, except for normal group, separately treated the rats through pouring into their stomach with distilled water, Colchicine liquor, three different dose ZBC decoction. The course of treat is 36 days. In the course of the experiment, we regularly observed the mental state, diet,hair,stool,urine and body weight of the rats.At thcend of the course of treat,we observed the surface, colour and lust, fabric of the rats' livers, computed the ratio of liver weight and body weight, spleen weight and body weight, and examined the serum liver function markcrs:alaninc amiotransferase(ALT), aspartate aminotransfcrasc(AST), aspartatc aminotransferasc-alanincamiotransferase ratio(ALT/AST), r-glutamyl transpcptidasc(GGT), total protein(TP), albumin(ALB), globulin(GLB) and albumin-globulinratio(A/G), serum fibrotic items: hyaluronic acid(HA), laminin(LN), type III procollagcn(PC III ) and type IV collagcn( IV -C), fibrotic items of liver tissue: hydroxyproline(IIyp), and mRNA expression of HI collagen. At the same time, we observed hepatic tissue pathological section of the rats with optical microscope and electronic microscope.Result: Through scientific statistics we analyzed the meaning of every item used in the experiment .The results showed that ZBC can improve the rats' mental state, diet, hair, increase their body weight,decrease remarkably ALT, AST, and GGT activity, raise ALB content and A/G, decrease the levelof serum HA, LN, PC III and IV-C, and decrease Hyp content of liver tissue, restrain mRNA expression of III collagen, relieve remarkably the inflammalion damage, fatly degeneration of the liver cells, and reduce the collagen fibre of the liver.Conclusion: ZBC is good at protecting liver cells, improving liver function, adjusting the metabolism of extracellular matrix(LCM), reversing hepatic tissue pathological change and reversing HF, it's mechanism is probably inhibiting the expression of gene. ZBC is a better complex prescription pharmaceutics on anti-hepatofibrosis.
