Objective:To study the expression of a novel inhibitor gene of apoptosis, survivin, in bladder transitional cell cancer, and its relationship with the expression of p53, bcl-2 and ki67 genes.Methods:Using streptavidin-biotin peroxidase(SP) method, we examined the expression of survivin, p53, bcl-2 and ki67 proteins in 58 bladder transitional cell cancer tissues and 10 normal bladder tissues.Results:Survivin was expressed in 41 of 58 (70.7%) cases of bladder transitional cell cancer. In contrast, normal bladder epithelium did not express Survivin. Overexpression of survivin was strongly related to the tumor grade and clinical stage. The rate of positive expression of p53,bcl-2 and ki67 was 72.4%,31.3%,58.6% in bladder transitional cell cancer. Positive expression p53,ki67 of was related to the tumor grade and clinical stage.The expression of survivin were strongly corelated with p53(34/42,81% versus 7/16, P<0.01), bcl-2(9/18 versus 32/40,80%, P<0.05) and ki67(28/32,82.5% versus 13/24, p<0.05) expression . Conclusion:1.Overexpression of Survivin indicate survivin paly a important role through inhibiting cancer cell apoptosis in bladder transitional cell cancer. Overexpression of Survivin indicates poor prognosis.2. Survivin may not only inhibit apoptosis but also promote proliferation. Apoptosis inhibition by survivin, alone or in cooperation with p53 and bcl-2 may participate in the onset and progression of bladder transitional cell cancer. Loss of bcl-2 expression, overexpression of survivin,p53 and ki67 is one characteristic of high grade bladder transitional cell cancer . Examining survivin,p53 and bcl-2 have reference value for development and prognosis in bladder tranditional cell cancer.3. Survivin could be a new diagnostic/therapeutic target in bladder tranditional cell cancer.
