ObjectiveIt is very important to know the relationship between VEGF and integrins . As knowen , in vivo, VEGF can upregulate the expression of v integrins ; but in vitro, the cultured endothelial cells are classified as microvascular and large vascular endothelial cells. This experiment was taken to determine the influence of vascular endothelial growth factor on the av integrin expression of large vascular endothelial cells in vitro, and to discuss its possible mechanisms to promote angiogenesis and wound repair.MethodsHuman umbilical vein endothelial cells were primarily cultured ,The experiment was classified as the control ( DMEM) group and the experiment (VEGF) group, the control group was affected with 1 ml DMEM without serum , the experiment group was affected with 1ml 20ng/ml VEGF165. Tne expression of av integrin was detected with SP immunocytochemical method.ResultHie expression of av integrin was on the HUVEC membrane, in the control groups, av integrin was weakly expressed; but in the experiment groups, av integrin was strongly expressed, compared to the control group, the mean gray value has significant difference (P <0.01 )ConclusionEctogenic VEGF can upregulate av integrin expression on human umbilical vein endothelial cells in vitro after 24 hours.
