Objective To detect HHV-6 and HHV-7 DNA in peripheral blood mononuclear cell (PBMC) and saliva specimens of healthy blood donors, to investigate the infection of HHV-6 and HHV-7 in Qingdao healthy adults.Methods HHV-6 and HHV-7 DNA in PBMC and saliva were tested by Nested-PCR, the amplified products of HHV-6 and HHV-7 were respectively digested by restriction enzyme Hind III and EcoR I to identify its specificity.Results (1)In PBMC and saliva specimens, the positive rate of HHV-6 infection is 87. 0%, and HHV-7 is 90. 6%. (2)The positive rate of HHV-6 infection in both PBMC and saliva is 33. 3%, HHV-7 is 58. 0%, and 78. 3% of blood donors were simultaneously positive for HHV-6 and HHV-7. (3)There is no difference of the positive rate of HHV-6 and HHV-7 infection among the sex, the age and the blood type, P>0. 05. (4)The difference of positive rate of HHV-6 and HHV-7 was significant between the internal and external reaction, P<0. 01.Conclusion The infection of HHV-6 and HHV-7 in healthy adults is common, and the positive rate is rather high. The blood donors infected by HHV-6 often have infection of HHV-7,the opportunity of infection of HHV-6 and HHV-7 in different sex, age and blood type individuals is equal. Nested-PCR combined with restriction enzyme technique is a sensitive, specific and effective method in detecting HHV-6 and HHV-7. There is a high positive rate of HHV-6 and HHV-7 infection in PBMC and saliva specimens of blood donors.
