| Honghua Injection(HHI) is the extract of Carthamus tinctorius L. At present HHI is widely used in treating many diseases such as coronary heart disease, cerebrovascular disease, the complication of diabetes mellitus and so on. The satisfactory therapeutical effects prompted us to explore the therapeutical characteristic and mechanism of HHI.The present experiments studied the effects of HHI on the blood system, especially on the aggregation of platelet, hemorheology and the formation of thrombus. The results showed that HHI 200mg/kg and 500mg/kg(ip) significantly prevented the aggregation of platelet induced by ADP in rats, the rate of prevention is 46.4% and 86.6% respectively. Experiments in vitro showed that HHI 8-16mg/ml significantly prevented the aggregation of platelet induced by ADP or collagen in rabbits. The research on hemorheology demonstrated that HHI markedly decreased the whole blood viscosity of rats, and there were no differences in plasma viscosity, hematocrit and erythrocyte sedimentation compared with before treatment. We prepared the carotid artery-cervical vein bypass according to the Umetsu's method. The research on thrombosis in rats showed that wet weight of thrombus lowered obviously when treated with HHI 100-500mg/kg(ip), and the suppression rate of thrombosis is 13.8-45.3%with markedly dose-effect relationship. The study of time-effect relationship indicated that the suppression effect of thrombosis can last at least 8 hours. We also observed the effect of HHI on thrombosis in rabbits in vitro applied the improved Chandler's method. The results demonstrated that HHI 200mg/kg and 500mg/kg iv significantly prohibited the forming of thrombus in rabbits in vitro including the phenomenon of the length of thrombus shorter, the weight lighten and the forming time longer.All the experiments showed that the effect of inhibiting platelet aggregation, decreasing blood viscosity and suppressing thrombosis is one of the mechanisms of the therapeutical action of HHI. |
