Oral squamous cell carcinoma(OSCC) is the most encounted in tongue gingiva, buccal mucosa, palate, and so on. Along with the development of cancer research, the traditional operation scheme have been changed in many ways, which makes the local control of OSCC become possible. In spite of combined with radiotherapy and chemotherapy, some cases neck-lymphonodi metastasis tend to result in treatment failure.The immunotherapy based on a great appreciation of the mechanisms by which tumor-specific cytolytic cells are generated is becoming a new approach in cancer treatment. Most human tumor are poor]y immunogenic and are capable of evading the host immune system. Accumulating evidence has demonstrated that tumor cells may down-regulate the expression of signals that are essential for the activation of host T cell. The mechanisms include defective expression tumor specific antigen(TSA) , tumor associated antigen(TAA) or MHC Ags, and alteration of Ag-processing or transport by APC, resulting in evading the host immune system. Several strategies of using TAA TSA or fusion with tumor cells as vaccines for cancer immunotherapy have been developed.Objective:1. To study the expression of main histocompatibility complex I II (MHC I II) in oral squamous cell carcinoma(OSCC) and its significance. 2. Try to employ a new immunotherapy strategy in this study by fusing entire Tca8113 cells with macrophages, in order to evaluate the therapeutic effectiveness of fusion tumor vaccine in OSCC treatment.Methods: OSCC were examined with immunohistochemical S-P technique for human leukocyte antigen ABC DR expression and analyzed the relationship between the expression and clinic stage neck-lymphonodi metastasis. We fused human macrophages with human tongue carcinoma cell line Tca8113. The biological properties of fusion cells and anti- tumor immune response in vitro induced by fusions were observed.Results:The expression of HLA-ABC, DR in OSCC were significantly variable among different pathological degrees(P<0.05). The expression of HLA-ABC DR in OSCC at clinic stage III IV was significantly lower than that at stage I , II. The expression of HLA-ABC in OSCC with lymphonodi metastasis was significantly lower than that without.In contrast to Tca8113, the fused cells grew significantly slow in vitro. The expression of MHC I II antigen which was detected by flow cytometry(FCM) was higher than Tca8113. The fused cells significantly increased the proliferation of mixed lymphocyte and induced thire cytotoxicity on parental Tca8113.The fusion tumor vaccine of macrophages and OSCC cells increase in vitro immunogenicity significantly. This indicatesthat fusion tumor vaccine could be new ways to induce effective anti-tumor immunotherapy, which have important potentials for effective individual human OSCC vaccine.
