OBJECTIVE: To study the gene expressions of Nestin and stem cell factor (SCF) after ischemic injury in rat brain, and to explore the therapeutic mechanism of inosine in central nervous system ischemic injury.METHODS: 68 adult female rats were divided randomly into treatment group and controlled group. The rats were received left middle cerebral artery occlusion 1.5h and different hours of reperfusion. Neurological grade evaluation and in site hybridization were used to examinate the behaviorcal recovery and expression of Nestin mRNA and SCF mRNA in the rats subjected to 2h, 6h, 12h, 24h, 2d, 3d, 7d, 14d of reperfusion and sham-operated group (n=4).RESULTS: (1) There was improvement in the score of neurologic grade in inosine-treated group compared with controlled group at reperfusion of 7d and 14d. (2) Nestin mRNA expression in cortex, striatum and extraventricular zone was weekly in sham-operated group. The expression of Nestin mRNA in ischemic hemisphere of controlled group increased markedly compared with sham-operated group (t=1.95~47.21, P<0.05), besides that of reperfusion 2h in cortex, 2h, 6h in striatum, 2h, 6h and 14d in extraventricular zone. Nestin mRNA expression in inosine-treated group decreased in cortex at reperfusion 2h, 6h, 7d, 14d and striatum at 14d compared with controlled group, then increased markedly in cortex, striatum and extraventricular zone at 3d, but increased only in striatum and extraventricular zone at 7d (t=4.37~18.03, P<0.05). (3) SCF mRNA expression in cortex, striatum and extraventricular zone was weekly in sham-operated group. The expression of SCF mRNA in ischemic hemisphere of controlled group increased markedly compared with sham- operated group (t=2.26~28.99, P<0.05), besides that of reperfusion 2h<6h< 12h in cortex, 2h,6h in striatum, 2h and 14d in extraventricular zone. SCF mRNA expression in treatment group decreased in cortex and striatum at reperfusion 14d compared with controlled group, then increased markedly in cortex, striatum and extraventricular zone at 3d and 7d (t=5.10-28.17, P<0.01).CONCLUSION: It is suggested that inosine could improve behavioral outcome of MCAO rats and promote the expressions of Nestin mRNA and SCF mRNA , so it might promote the creation of neural stem cell.
