Objective:We did lots of experiments to research the feasibility and method of tissue microarray so that we could provide a fast, simple and economical technique for scientific study and clinicopathological diagnosis.Material and Method:Donor blocks including 54 lung cancer and 10 normal lung tissues. Sections cutting from the blocks was 5 u m each and stained by HE method. We marked the needed tissues on the donor blocks with the help of HE slides and collected all of the tissue cores into the recipient blocks in terms of the previous design (20 X 12). We selected 4 tissue cores from each lung cancer cases and 2 from normal lung samples. After staining sections which were 3 u m each by HE method, we detected these sections with anti-EphB4 poly-antibody by SP immunohistochemical technique. Of the 54 cases of lung cancer, We compared the staining of 1 to 4 array cores and the whole tissue sections from which they were derived and determined the coincidence rates between them each other.Results:1. Facture of tissue microarray: The character of our recipient block was exquisite and had no splits. Disks were arranged clearly on the slides and had no shift and distort. They were answered for the request of research.2. Accord rate between the anti-EphB4 poly-antibody staining of 1 to 4 array cores and the whole tissue sections from which they were derived: Tissue microarray analysis showed the accord rates were respectively 96.3% (52/54, 4 cores examined), 94.4% (51/54, 3 or 2 cores examined) and 90.7% (49/54, 1 core examined) . There were no significant differences between them each other and whole section (p>0. 05) .Conclusions:1. Our tissue microarray technique was successful, and it could be used on research work and clinicopathological diagnosis.2. Tissue microarray is a new technique which has characters of high-throughout, economy and low rate of experimental error.3. When compounding tissue microarray technique to immunohistochemical method, taking 2 cores from the whole-tissue sections was an accurate and ecomomicai method.
