Objective: Nonalcoholic fatty liver disease(NAFLD) is a clinical syndrome characterized by excessive fat storing in liver and hepatic cellular degeneration,without alcohol over ingestion .The incidence of fatty liver is increasing and NAFLD patients are prone to liver sclerosis or liver failure. So it is necessary to find a new way on therapy of fatty liver.Methods: Male Wistar rats were divided randomly into five groups:normal control group(group N),was fed with normal diet,model group(group M),large dose Chinese herbs group(group H),small dose Chinese herbs group(group L)and Gemfibrozil group(group G)were fed with high fat dietAfter 4 weeks,group H,group L and group G were perfusated with Chinese herbs and Gemfibrozil respectively for 8 weeks.group N and group M were perfusated with lml/lOOg NS. Pathological Changs of liver, serum levels of TC, TG, ALT, AST, FFA, SOD, MDA, liver levels of TC, TG, FFA, SOD, MDA and the expression of hepatocyte cytochrome P450II El were observed.Result:We established the NAFLD rat model successfully. Diffuse macrovesicular steatosis mixed with inflammatory cell infiltration was showed in livers of group M.Compared with group N, group M had higher levels of TC, TG, FFA, ALT, AST, MDA and lower level of SOD and increased expression of CYP2El,all the above results had statistical significance(P<0.05).Contents of TC, TG and FFA were decreased in group H than in group M(P<0.05),although the levels of them were higher than in group N; MDA and CYP2E1 were decreased and SOD activities were increased in group H than in group M(P<0.05).The theraputic effect of large dose Chinese herbs was similar to that of Gemfibrozil .while Gemfibrozil could increase aminotransferase activity. Conclusion:In group M, weight, liver weight, serum lipid, liver lipid, aminotransferase and lipid peroxide products-MDA were increased and hepaticcellular macrovesicular steatosis was showed histologically;the incidence of fatty liver in rats with high fat diet was 100%.Chinese herbs not only decreased levels of serum lipid,liver lipid,lipid peroxide products but also protected hepatocyte from injury.In a word,these results showed Chinese herbs has strong and safe effect on therapy of nonalcoholic steatohepatitis.
