Objectives To explore the characteristics of hepatitis B virus genotypes in the subjects with chronic HBV infection. Methods HBV DNA levels and genotypes were determined by quantitative fluorescent PCR and nested PCR using type-specific primers,respectively. Result:1. The sensitivity of nested PCR using type-specific primers was 102 copies/ml2. Among the patients with chronic hepatitis B (CHB) or liver cirrhosis (LC), the positive rate of genotyping genotype were 68.3%(28/41), 88.4%(38/43) and 99.1%(96/97) respectively when HBV DNA loads in the sera were <103, between 103 and 105and >105 copies/ml (p<0.05. 3. The positive rate in the HBeAg positive samples was significant higher than that in the patients without HBeAg(91.8% vs 82.8%,147/158vs250/302, X2=9.232, p=0.002).4. 236 of 395 cases in positive genotyping (59.7%) were genotype B, 97 (24.68%) C, 62 (15.7%) B and C coninfections. 5. Genotype B was significantly more prevalent in ASC (asymptomatic HBsAg carrier), CHB and CHF( chronic fulminant hepatitis B patients), and the same as genotype C in LC group and genotype B and C coinfection in HCC( hepatocellular carcinoma )group.6. The HBV DNA loads in the sera were significant higher in the patients with genotype B infection than those with genotype C infection.Conclusion Nested PCR using (A-F) type-specific primers is highly sensitive, simple and fast, and suitable for clinical routine use. Genotype B is the predominant genotype in Fuzhou city. Genotype C or B and C coinfection might have a close relationship with the serious liver diseases such as LC and HCC.
