[Objective]1 To establish the model of allergic rhinitis(AR) of SD rat and observe the histologic change in nasal mucosa. 2 To study the expression of SP and VIP in nasal mucosa of AR and investigate the relation between SP and VIP with AR.[Methods] 50 healthy SD rats were devided into the normal,negative control and AR groups including 30 miniter after stimulate(early phase response, EPR)and 9 hours after stimulate(late phase response,LPR) respectively. After establishing the model of allergic rhinitis by intraperitoneal injecting comphensive solution of ovalbumin(OV) and AL(OH)3 then droping 5% OV solution to nose. Apply transmission electrical microscope to analysis of ultrastructure and hisochemical technique to study histologic change innasal mucosa of AR,such as the master cell ,the subepithelial glands et al. Expression and varition of SP and VIP in the nasal mucosa of allergic rhinitis were observed with immunohistochemistry technique (SABC method).[Results] there were loss and down of the cilia ,increasing of the master cell ,lympha cell and EOS, hyperplasia of basal cells, and increased height of the cylindric cell in different degree in nasal mucosa of AR. Although the nosal mucosa of nomal and negative control groups shown the much typical appearance of a pseudostraified columnar ciliated epithelium or basal layer, without cilia loss and increasing of inflammatory cell. All groups were positive for SP and VIP, Positive SP and VIP terminals were located along the walls of small vessels, around the glands and between the epithelial cells of nasal mucosa. but SP and VIP terminals in AR were denser than nomal and negative control groups significantly.[conclusion] The development of EPR of AR allary comply with LPR of AR. They both have same change of pathology,such as despair of cilic, increasing of glands and secretion, welling of small vascation. The master cell and EOS were increased in AR group, in EPR and LPR there were different in quality and reaction in master cell and EOS, which may have different faction in EPR and LPR of AR. There were exprssion of VIP and SP in nomal nasal mucosa. But they were significantly increased in nasal mucosa of AR. they may imposed the development of AR.
