According to the related requirements issued by the State Food and Drug Administration (SFDA), the extraction and isolation technologies of polysaccharides from Herba Spirodelae seu Lemnae (PHS) were explored in this paper. The purity of PHS satisfied the application requirements (about the fifth class of herbal medicine) of SFDA. To test the immunomodulatory action of PHS, preliminary studies on PHS's pharmacodynamics were carried out. The main results were as follows:Polysaccharides, the immune-active component, were extracted and isolated subsequently by decoction, ultrafiltration, removal of proteins, and ethanol precipitation et al. And the processing technology using ultrafiltration was explored, which we'd like to emphasize. The manipulation condition of ultrafiltration was moderation, which would not cause the decomposition of PHS. The equipments of ultrafiltration were simpleness; the operations were convenient and with no substance shifted. The condition controlling in the removal of protein by trichloroacetic acid (TCA) was very rigorous, but it did be very effective. After precipitating the polysaccharide with ethanol for 6 times, the absorbency of the sample increased significantly through sulphuric acid – phenol testing. Besides, the pigment in the extraction has been got rid of partly. The purity, the contents of protein, and the recovery were 53.5%, 1.2%, and 0.08%, respectively, in the last obtained hazel powder. (Table 2.2).It offered a significant reference in establishing the quality standard of PHS in this paper for the applied steps and methods, as follows. First, the saccharides content was mensurated by spertrophotometry of sulphuric acid – phenol and sulphuric acid – anthrone. Then the methods were evaluated through the indexes as linearity and range, precision, stability, and accuracy and so on. The results indicated that the methods were convenient, rapid, accurate, high sensitive and could well be used to determine the saccharides contents of PHS. Lastly, the effects of PHS on phagocytosis of macrophage and mononuclear phagocyte system were investigated. PHS was shown to enhance the function of non-specific immune response obviously (Table 4.1, Table 4.2). In brief, a simple, quick and effective method to extract and isolate polysaccharide from Herba Spirodelae seu Lemnae was established aiming at increasing the purity of PHS extraction. So that the standard on the application of new medicament required by SFDA could be achieved. Meanwhile, the method of testing total saccharide content in PHS and how to evaluate it were investigated. The immunomodulatory activity testing offered much help to the medical study and application of PHS.
