Aim: It's proved that a Chinese tranditional medicine,Huangqi(Astragalus),can inhibit apoptosis in some kinds of cells.The experiment was designed to investigate if astragalus can inhibit cryptorchidism-induced apoptosis in germ cells,further more alleviate the pathological changes in undescended testes.Method:26 immature male Wistar rats was devidied into two groups equally,all two groups were established into unilateral cryptorchidism animal model according Dundar Mafter's method.After operation,experiment group received injection of astragalus for one week, control group received same dose of physiological saline for one week.and then all animals were killed at the 8th day after the surgery. All the testes were removed,decapsulated and weighted,the pathological changes were studied under light microspope with HE slices, the apoptotic germ cells in bilateral testis were studied with terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick and labeling.Results: The weight is different between the ipsilateral undescended testes(UDTs) and the contralateral descended testes(CDTs) in both groups,the decreased weight of UDTs in experiment group is significantly lower as compared to the control group. The density of seminiferous tubules decreased and the seminiferous tubular diameters were significantly reduced in the UDTs in both groups,but the impairment in experiment group is lighter in the comparison with the control group. the apoptotic index of germ cells in experiment group is lower than the control group (P<0.05).Conclutions:Cryptorchism induces pathological changes in undescended testes,which inculding the descease of the weight, the decrease of the density and the diameters of seminiferous tubules. UDTs demonstrated a markedly increased incidence of apoptosis in both groups, Astragalus can mitigate the pathological impairment and inhibit the apoptosis in germ cells of undescended testes.
