Objective: To investigate the effect and mechanism of Foeniculum Vulgare Mill on liver fibrosis in rats. Methods: 60 healthy Wistar rats were divided into six groups randomly: group A is normal control group, feed without any drug. Experimental hepatic fibrosis was induced in groups B,C,D,E and F as Han Dewu reported. In B,C and D groups, 8 rats in each group were killed at the 4th weekend. At the 8th weekend, all rats were killed, livers were collected for HE,Masson stain, Serum was collected for the detection of ALT,TP,ALB,Hyp and MDA. Results: In group A, morphological and biochemical test were normal. After 4 weeks of experiment: (1) Livers in each group are swollen, the color of the liver become brown. In group B, some liver's border become blunt, there are conglutination between lobes or lobes and peritoneum. (2) HE stain show inflammation is diffuse in group B, the fatty degeneration of hepatocyte is obviously, there is plenty of inflammatory cells in portal tracts and around central veins while the inflammation and the fatty degeneration of hepatocyte are feeble in group C and D (p<0.05). (3) Masson stain show that relative majority portal tracts are expand and full of collagen fiber in group B, and the fibrous septa encircled the hepatic lobule, but the lobular structure is normal. In group C and D, the fibrosis is lighter (p<0.05) and the expansion of the portal tracts is not so seriouscompared with group B. (4) The biochemical tests show that ALT in all experimental groups is markedly increased than group A(p<0.01); TP and ALB are decreased, the difference between group A and B has statistically significant(p<0.05,p<0.01), the difference of TP between group A and D, ALB between group B and D are markedly(p<0.05, p<0.01). MDA in group B is markedly increased than group A(p<0.01), in contrast, in group C and D it is decreased, but the change is inconsequential. Hyp in group B,C and D is markedly increased than group A(p<0.01)and group C and D is markedly higher than group B(p < 0.01). After 8 weeks of experiment: (1) Livers in group B is dwindled obviously, the surface is rough, its color is brown, the border is blunt, 4 rats have ascites, the capacity is from 20 to 30 ml; and it is slightly swollen in group C and D while it is slightly dwindled in group E and F. (2) HE stain show that fatty degeneration and inflammation are light but pseudolobe is more obvious than the end of 4 weeks in group B; in group C,D,E and F, inflammation is feeble and pseudolobe can be seen in some areas compared with group B, (3) Masson stain show that collagen fiber hyperplasia and the cirrhosis are obviously in group B while both of them are slightly in group C,D,E and F(p<0.05). (4) The biochemical test show that ALT is markedly higher than group A in group B,C,D,E and F(p < 0.05), in group C and F, it is lower than group B(p < 0.01); TP in group B,C,D,E and F are markedly decreased than group A(p < 0.05), in group C,D, E and F, it is higher than group B, the difference between group D,F and B is markedly (p < 0.05); ALB in group B,C,E and F is markedly decreased than group A(p < 0.05) except group D, the change between group B,C,D,E and F is inconsequential (p > 0.05); MDA in group B is markedly increased than group A (p < 0.01), in group C,D,E and F, it is decreased and the change is inconsequential compared with group A but markedly with group B; Hyp in group B,C,D,E and F is markedly increased than group A(p < 0.01), in group C,D,E and F, it is markedlyhigher than group B(p < 0.01). Conclusion: Foeniculum Vulgare Mill can inhibits the inflammation in rat liver and protects the hepatic cells from damage induced by CCL4; it can accelerate the collagen dissolution in the fibrous liver and reverse the hepatic fibrosis; the mechanism is related to the effect of Foeniculum Vulgare Mill inhibits the lipid peroxidation in liver and the activation and proliferation of the hepatic stellate cells...
