Objective:To observe the effects of Foeniculum Vulgare Mill on liver fibrosis and to explore its mechanism related to hepatic lipid peroxidation in rats. Methods:Liver fibrosis model was established in healthy Wistar rats by the combination of injecting carbon tetrachloride (CCL4) subcutaneously and feeding high lipid diet and drinking 10% alcohol. The model rats were randomly divided into 5 groups:the model group (n=12), Foeniculum vulgare mill therapeia group (n=12/group) (low, middle, high), danshen group (n=12), Besides, a normal group was set up with 12 normal rats; Drugs were lavaged for 8 weeks. For the treated group, rats were administered with Foeniculum Vulgare Mill at a dosage of 0.54 g/kg body weight,1.08 g/kg body weight,2.16 g/kg body weight,1.02 g/kg body weight once a day by gastrogavage starting from the day after modeling for 8 successive weeks and to the control group, equal volume of normal saline was administered instead. At the 16th weekend,all rats were killed, The inflammation and fatty degeneration in rat liver tissues were examined with HE staining;the liver function including serum level of alanine transaminase(ALT),aspartate aminotransferase (AST), albumin(AlB)and total protern (TP) were determined using corresponding test kits;the hepatic lipid peroxidation indexes,including malondialdehyde(MDA), superoxide dismutase (SOD), Glutathione peroxidase (GSH-PX),Catalase (CAT) were measured by bioche nical methods; Results:Compared with those in the normal rats,serum ALT,AST level and MDA content remarkablely increased,level of TP,ALB and SOD,GSH-PX,CAT activity decreased,and hepatic fatty degeneration and collagen pathological deposition in liver was more obvious in the model rats(all P<0.05).While in the Foeniculum Vulgare Mill group,serum ALT,AST level and MDA content remarkablely decreased,level of TP,ALB and SOD,GSH-PX,CAT remarkablely increased,and hepatic fatty degeneration and collagen pathological deposition in liver was more obvious in the model rats(all P<0.05). Conclusion:Foeniculum Vulgare Mill can inhibits the inflammation in rat liver and protects the hepatic cells fromdamage induced by CCL4; Foeniculum Vulgare Mill it can accelerate the collagen dissolution in thefibrous liver and reverse the hepatic fibrosis; the mechanism is related to the effect of Foeniculum Vulgare Mill inhibits the lipid peroxidation inliver and the activation and proliferation of the hepatic stellate cells.
