| Objective: To observe the immune mechanism of rejection of chemical extracted acellular allogeneic nerve for further use in clinic.Methods: In the first part of our experiment,we initially obtained acellular nerve allografts including chemical extracted acellular allogeneic and xenogeneic nerve grafts.The grafts were excised from sciatic nerves of 20 C57BL/6 mice and ulner nerves of 7 rabbits(1.5cm in length,0.3cm in diameter) and treated with a solution of Triton X-100 and a solution of sodium deoxycholate. Then we used 2 chemical extracted acellular xenogeneic nerves and 2 fresh xenogeneic nerves of rabbit for morphological and immunohistochemical studies.The chemical extracted acellular nerve of rests were preserved for use of the second part of the experiment.In the second part of the experiment, 192 BALB/C mice were random divided into 6 groups, including negative contrast group(32 cases); fresh autograft group(32 cases); fresh allogeneic nerve group(32 cases); fresh xenogeneic nerve group(32 cases) ; chemical extracted acellular allogeneic nerve group(32 cases) and chemical extracted acellular xenogeneic nerve group(32 cases). Then we put various kind of nerve grafts into the thigh muscle of 192 BALB/C mice in corresponding groups. By the time of 3,7,14,28 day postoperative, the mice(each group 8cases) were killed, and the spleens were picked out. Spleens were grinded into paste, erythrocyte and other cells were burst by distilled water, and T lymphocytes were collected up.
