There are two parts in the studies . The first one was the study on anti-tumor activity of Xiao Ai Gen(XAG),in this part,it was necessary to make toxicological assessment firstly,because XAG is kind of folk TCM drug which used in HuaZhou Area, GuangDong province,then we studied on anti-tumor activities of XAG through method of Serum Parmacology.The other one was studied on anti-tumor activitiese of Dan-shen Parenteral Solution (DPS)and evodiamine(EVO). Similarly,the first step is make sure there are growth(proliferation) inhibition effects of dan-shen parenteral solution and evodiamine on H22 cell.the further research was studied dan-shen parenteral solution and evodiamine might induce apoptosis on H22 cells.Methods: (1).Using tests of acute oral toxicityon Kunming mice;(2).By means of Serum Pharmacology, XAG acted on H22 cells,then observing effects of growth(proliferation) inhibition on H22 cells,and also effect of apoptosis by FCM;(3).The growth inhibition of dan-shen parenteral solution and evodiamine on H22 cells were measured using MTT array;then calculate IC50 of the DPS and EVO;(4).The apoptosis of dan-shen parenteral solution and evodiamine on H22 cells were measured using DNA agrose gel electrophoresis and flow cytometry, the cellular changes was observed by microscope. (5) .All datas were analyzed by statistical software SPSS12.0,comparison between groups using method of LSD.Results: (1).The maximum dosage of XiaoAiGen(XAG) on Kunming mice was 17.5g/kg.d,the dosage correspond tol05 times (> 100 times) of clinic use,no death;(2). XAG could obviously suppress the proliferation of H22 cells.The survival rate(%) of H22 cells after cultrure 48 hours with serum of XAG as follow, 25% control group: 100±13.1, serum of higher dosage XAG 25%group: 83.2±10.5, 50% control group 100±22.3, serum of higher dosage XAG 50%group: 67.2±9.8.It was found that the higher dosage serum of...
