Objective: Erythropoietin(EPO) is a glycoprotein hormone response to tissue hypoxia. Lately, people find EPO is present in some tumors and also an effective cytokine in promoting angiogenesis. To explore (1)whether the EPO is present in gastric carcinoma ; (2)the possible mechanism of its expression; (3) the role of EPO in tumor angiogenesis and its results, we investigated the expression of EPO, HIF-la in gastric carcinoma and marked EC with CD105( monoclonal antibody MS1290 )and CD34 on paraffinic slides. Analysis of the realationship between EPO MVDand HIF-la in human gastric carcinoma.Methods: 82 cases of gastric carcinoma and the tissues nearby were detected on Tissue Microarries which was made by Tissue Arrayer. EPO ,HIF-la,CD105 and CD34 were visualized by immunohistochemistry SP. The results were tested by statistics methods(t-test chi-square test Spearman' rank correlation).Results: (1)The ratio of over-expressed of EPO in gastric carcinoma is 70.0% while the tissues nearby is almost negative. The distinction between the two sites is marked ( P<0.01 ) ;(2)The expression of MVD increased significantly with increased expression of EPO and HIF-la(P<0.05). The MVD of EPO(+) was much higher than that of (-);The MVD of HIF- la(+) was higher than that of (-).The level of EPO expression was closely related with MVD level(r=0.267, P<0.05).(3)The percentage of over-expressed cases of EPO was associated with the condition of the diatant metastasis (P<0.05). In PTNM staging of gastric carcinoma,the percentage of over-expressed cases of EPO has no different between I and II ,but the distinction between I II and III or IV had statistical significance (X~2=6.31,P<0.05; X~2=6.88,P<0.01). (4)The expression of EPO shows positive closely correlation with the expression of HIF-la (P<0.01).(5)The relationship between the two groups of MVD counting by CD105 and CD34 staining had significant difference (P<0.001).Conclusion: (1) EPO was expressed in gastric carcinoma, the ratio of over-expressed is high. (2)EPO has a closely relationship with angiogenesis in gastric carcinoma. It act as an angiogenic growth factor which can affect the biological behavior of tumors.3)EPO and HEF-la are closely interrelated, it maybe a new pathway in tumor angiogenesis through hypoxia. Blocking the pathway may help us in curing tumor.(4)The MS-1290 has high affinity with newborn blood vessels on paraffin sections and can reduce the false-positive errors in the study of tumor angiogenesis.
