BACKGROUND AND OBJECTIVEBronchial asthma is a chronic airway inflammation which associated with lymphocyte eosinophil mast cell monocyte and other cells and cellular component that widely participate.NF- K B is a multiple function transcription protein that widely exist.lt participate in regulating the express of lots of cytokines and inflammation media,and is closely associated with inflammatory disease.lt is the key transcription factor which associated with the expression of lots of inflammatory media in the pathological process of inflammation.lt play a center role in the inflammation process of onset enlargment and last. The balance of Th1/Th2 play an important role in the immune regulation.The patients with bronchial asthma exist the predominant coverting or migrant from Th1 to Th2.The predominant Th2 reaction play an important role in the pathogenic machanism of asthma. IL-4 and IL-5 produced by Th2 is regarded as the key cytokines.IL-4 is the characteristic cytokine of Th2, which can selective induce and promote the production of IGE.IL-5 can promote the collection infiltration and activation of EOS.IFN- Y is the characteristic cytokines of Th|. It caneffectively active the mono-megacaryocyte,and promote its fight ability.IFN- γ and IL-4 is a couple of cytokine that can not only agonist each other but also promote respectively.They are the represent of Th1 and Th2 respectively.Thymosin-α ,as a immune regulation chemical,can maintain the body in a station of immune balance.It can promote the lymphcyte to produce IL-2 and IFN-γ .The patients with asthma exist the disorder of the Th1/Th2 balance,and exist migration from Th1 to Th2.T0 suppose on theory, Thymosine- α have the important therapy value on asthma.Dexamethasone is an important immune inhibitor,which is a long-effect glucocorticoid.As a kind of classic medicine to therapy asthma,it is very effective.At present it is still the first choice to therapy asthma.The aim of the study is to observe the expression of NF- k B and Th1/Th2 cytokines in asthmic mice model and try to find out their function in the pathogenic process of asthma.,and under the circumstance of thymosine- α and dexamethasone interfering with,the change of each of the involving factor above and their clinical utilizing price.MATERIAL AND METHODS1 .Twenty-eight male BALB/C mice was bought.The laborory animal was SPF grade. They are divided into 4 groups evenly,which are normal control group,asthmic model group,and thymosine- α group,dexamethasone group.2.At the beginning days,the seventh days and the fourteenth days,the normal group was injected with NS 0.5 ML into peritoneum,while the asthmic model and thymosin - α group dexamethasone group was injected with mix fluid 0.5ml(2mg AL(OH)3 and 10 μ G OVA) respectively in order to be sensitived.In addition,the thymosin - α group was injected in the way of QOD from the beginning day with thymosin -α .All the animal were stimulated with 2% OVA solution delivered by aerosolization on days 15 16 and 17.Dexamethasone group was interfered by Dex aerosolization on days 15 16 and 17 before it was stimulated.3.At the eighteenth day,we collect the BALF.After that we take out the bronchial-lung tissue.4.The level of IL-4 IL-5 and IFN- γ of BALF was detected by ELISA. The activity of NF- k B was detected by immunohistochemical staining.5.Comparing the level of cytokines and the percentage of NF- k B of each group,and make use of the SAS system to undertake the statistic analysis. RESULTl.WBC counts and classification in BALFComparing the counts of EOS and lymphocyte between the normal group and the asthmic group,it has a significant difference(p<0.05).Comparing the level of EOS of asthmic group to that of T- α group,it has significant difference(P<0.05). Comparing the lymphoctye level of normal group to that of T- α group,it has significant difference(P<0.05).
