| Objective: A long time ago, repair of peripheral nerve is still a difficulty. Peripheral nerve autograft is the best method of repair a long nerve defection in clinic. But the donar nerve is limited and results in certain function obstacle. Besides,it is anther problem that nerve growing speed is 1mm/d after nerve injury.All experts in the world have done much to solve the two problems. Recently,many experts pay their attentions on allografting.The nerve allograft source is wide and acquires easily,having the vast clinical application foreground.Successful nerve allografting mainly be decided by how to relieve immunological rejection between allograft and host. Several methods of pretreatment of nerve allograft to lower its antigen contain chemistry method, refrigeration method, dry method, irradiation and so on. Another method is the immunity depressant, prednisone, or applied at the same time all to obtain better result. It is an important factor that the speed of regeration is 1mm/d after injury of nerve, it will be affect the curative effect. If the muscle is out of contral of nerve injury a long period, the muscle fiber appears to atrophy, disvolution, fibrosis, losing the possibility of function instauration thoroughly. Therefore,it is the aim for clinical nerve studying worker to accelerate the peripheral nerve regeneration speed from 1mm/d to 2-3mm/d.FK506 is separate from the streptomyces, a kind of antibiotics of macrolide. Tacrolimus, its English name, often is translated to Te Ke Lei Mu or Te Ke Mu Si. His merchandise name is Pu Rui BO Si, with FK506 for the laboratory. The main function of FK506(1) represses the rejection of organ transplants (2) accelerate the peripheral nerve regeneration.The clinical allograft of hand found that FK506 can accelerate the regeneration of peripheral nerve speed to 3 mm/ d.There are two dosage form:phleb-prep,containing two kinds of 5mgs ,10mgs/ml,commenly are used through the physiology brine or the glucose liquid admixtures the vein drop note; take orally the product contain two kinds of 1 mg and 5 mgs.The main adverse reaction that commenly occurres at the vein way, have the headache familiarly, disgusted, vomit, insomnia and central nervous system system symptom tremor; a handful of patients still have the hot feeling,or suffer from the hypomagnesemia,moreover,the FK506 can lead to the blood sugar influence for period.How to decrease the side effect coming from medicine,availably work in the segment of allograft,ease rejection,and promote the growing at the same time.The experiment design:(1)cold preservation of nerve allograft regeneration with implantation of slow-releasing diaphragm with FK506 whether surpasses fresh allograft ornot;(2) cold preservation of nerve allograft regeneration with implantation of slow-releasing diaphragm with FK506 whether surpasses cold preservation of nerve allograft or not;(3) cold preservation of nerve allograft regeneration with implantation of slow-releasing diaphragm with FK506 whether differes from fresh nerve autograft.According to this ,we design this experiment.Come to investigate a method of allograftion in an a kind of better proceeding nerve regeneration. Methods:60 adult male SD mices,each weighing between 150 to 200g, is divided into 4 groups of 15 animals randomly.Group A:fresh allograft.Group B:cold preserved sciatic nerve allograft .GroupC:cold preserved nerve allograft receiving slow-releasing of FK506 from the diaphragm.Group D: isograft.Another 24 adult male Wistar mices ,are served as nerve donors.The two sides sciatic nerves were cut off,15 of them were grafted into group A and the others were put into three kinds of glycerin by turns for 3 hours each time,whose concentration was 50%,70%,85%, then stored in the glycerin closeness bottle whose concentration was 85% and tempratrue was 5℃for 1 week, which are for the groups B,C.The nerve need be put into the saline for 30 minutes before allograftion.Each allograft is sutured with the defective sciatic nerve by 11-0 nylon,and put the diaphragm of FK506 only into the...
