| Objective : To investigate the regeneration of peripheral nerve allografts stored in the glycerol of different time.Methods: 72 male SD rats in this experiment were carried out randomly in 6 groups. In group A, the grafts were fresh allografts. In group B, the grafts were fresh autografts. In 3 WEEK group, the allografts were stored in glycerol for 3 weeks. In 5 WEEK group, the allografts were stored in glycerol for 5 weeks. .In 7 WEEK group, the allografts were stored in glycerol for 7 weeks .In 9 WEEK group, the allografts were stored in glycerol for 9 weeks . Other 24 male SD rats were used for supplying the 48 sciatic nerves. After the storage , the allografts were transplanted to the corresponding rats. 2 weeks later, two grafts of transplanted nerves were harvested randomly and examined with immunohistochemistry. 9 and 16weeks later, the regeneration of sciatic nerve was examined with histology, electro-physiology.Result: Wallerian degeneration was observed in the proximal and distal ends of all grafts of nerves harvested at 2 weeks post-engraftment.There were so many T lymphocyte in group A. 3 and 5 WEEK group is less.none can be seen in 7 and 9 WEEK group. 9 and 16weeks after the operation , all of the allografts were adhering to the surrounding tissues. Group A was the most evident. Group 3 WEEKS was inferior, the rest won't evident. There were a lot of collagenoblast and lymphocyte in group A. The Wallerian degeneration is manifest and the nerve regeneration is worst .Otherwise, the 9 WEEKS group shows little collagenoblast and lymphocyte.The nerve regenerated obviously.The 3,5,7 WEEK group is between them.Conclusion:1.To increase the storage time can decrease immunogenicity.2.The best storage time is 9 weeks. |
