Objective: The purpose of this study was to investigate the expression of p53 protein and vascular endothelial growth (VEGF), and microvessel density(MVD) measurement in esophageal squamous cell carcinoma tissue and 2.0cm,5.0cm adjacent noncancerous tissue from esophageal squamous cell carcinoma to evaluate the relationship between these factors. In order to explore the role and clinipathological significance of p53 protein, vascular endothelialgrowth factor expression and microvessel density in esophageal cell squamous carcinoma tissue. Methods: Thirty samples of paraffin sections of esophagus squamous cell carcinoma were selected. The S-P immunohistochemical method was used to determine the expression of p53 protein and VEGF, Microvascular density was calculated, and vascular endothelial cells were labeled by Factor ⅧRlated Antigen (FⅧ).To detect the relationship between the expression of p53 protein, VEGF, MVD and differerent degree, clinical stages,lymph node metastasis, sex and age. All the data for statistics using SPSS10.0. Results: 1 The expression of VEGF: ①The positive staining of VEGF was brown, and mainly located In cytoplasm. ②The positive rate of VEGF was 73.3%(22/30) in esophageal carcinoma tissue.The positive rate of VEGF in 2.0cm,5.0cm adjacent noncancerous tissue from esophageal squamous cell carcinoma tissue were 33.3%(10/30), 26.7%(8/30)respectively. Statistic analysis showed that the positive expression rate had siginicantly different in different tissue(χ~2=15.480, p=0.001). ③The positive rate of VEGF were 51.8%(14/20) in male, 80.0%(8/10) in female. Statistic analysis showed that the positive expression in esophageal squamous cell carcinoma tissue rate had no siginicantly different in male and female (p=0.682). ④The positive rate of VEGF in esophageal squamous cell carcinoma tissue were 60.0%(9/15) in >55 years, 86.7%(13/15) in ≤55 years. Statistic analysis showed that the positive expression rate had no siginicantly different in >55 years and ≤55 years (p=0.215). ⑤The positive rate of VEGF in different TNM stages in esophageal cell carcinoma tissue were 56.3%(9/16)in Ⅰ,Ⅱstages, 92.3%(13/14) in Ⅲstage. Statistic analysis showed that the positive expression rate had siginicantly different in different TNM stages (p=0.039). ⑥The positive rate of VEGF in esophageal cell carcinoma tissue were 88.2%(15/17), 53.8%(7/13) with or without lymph node metastasis. Statistic analysis showed that the positive expression rate had siginicantly different in esophageal squamous cell carcinoma tissue with or without lymph node metastasis (p=0.049). ⑦The positive rate of VEGF in esophagealsquamous cell carcinoma tissue were 85.7%(18/21) in ≤5 years, 44.4%(4/9) in >5 years. Statistic analysis showed that the positive expression rate had siginicantly different in different lifetime in esophageal squamous cell carcinoma tissue (p=0.032). 2 The expression of p53 protein: ①The positive staining of p53 protein was brown, and mainly located In nucleus. ②The positive rate of p53 protein was 62.2%(19/30) in esophageal squamous cell carcinoma tissue. The positive rate of p53 protein in 2.0cm, 5.0cm adjacent noncancerous tissue from esophageal squamous cell carcinoma tissue were 23.3%(7/30), 23.3%(7/30) respectively. Statistic analysis showed that the positive expression rate had siginicantly different in different tissue( χ2=13.870, p=0.001). ③The positive rate of p53 protein were 60.0%(12/20) in male, 70.0%(7/10) in female.Statistic analysis showed that the positive expression in esophageal squamous cell carcinoma tissue rate had no siginicantly different in male and female(p=0.702). ④The positive rate of p53 protein in esophageal carcinoma tissue were 60.0%(9/15) in >55 years, 66.7%(10/15) in ≤55 years. Statistic analysis showed that the positive expression rate had no siginicantly different in >55 years and ≤55 years (p=1.000). ⑤The positive rate of p53 protein in different TNM stages in esophageal squamous cell carcinoma tissue were 62.5%(10/16) in Ⅰ, Ⅱstages, 64.3%(9/14) in Ⅲstage. Statistic analysis showed that the positive expression rate had no siginicantly different in different TNM stages (p=1.000). ⑥Thepositive rate of p53 protein in esophageal squmous cell carcinoma tissue were 82.4%(14/17), 38.5%(5/13) with or without lymph node metastasis. Statistic analysis showed that the positive expression rate had siginicantly different in esophageal squamous cell carcinoma tissue with or without lymph node metastasis (p=0.023). ⑦The positive rate of p53 protein in esophageal squamous cell carcinoma tissue were 76.2%(16/21) in ≤5 years, 33.3%(3/9) in >5 years. Statistic analysis showed that the positive expression rate had siginicantly different in different lifetime in esophageal cell carcinoma tissue (p=0.042). 3 The expression of MVD: ①The positive staining of MVD was brown. The distribution of positive cells was frequently observed at the Invading edge of tumor margins. ②The mean of MVD was 34.07±3.91 in esophageal squamous cell carcinoma tissue. The mean of MVD in 2.0cm, 5.0cm adjacent noncancerous tissue from esophageal squamous cell carcinoma tissue were 19.95±2.68,19.05±2.69 respectively. Statistic analysis showed that MVD in esophageal squamous cell carcinoma tissue and 2.0cm,5.0cm to esophageal squamous cell carcinoma tissue had siginicantly different (F=25.01,p=0.0001). ③The mean of MVD were 33.50±4.63 in male,35.20±7.49 in female.Statistic analysis showed that the MVD in esophageal squamous cell carcinoma tissue had no siginicantly different in male and female (T=0.396,p=0.695). ④The mean of MVD in esophageal squamous cell carcinoma tissue were33.64±6.43 in >55 years, 32.43±5.41 in ≤55 years.Statistic analysis showed that the MVD had no siginicantly different in >55 years and ≤55 years (T=0.283, p=0.779). ⑤The mean of MVD in different TNM stages in esophageal squamous cell carcinoma tissue were 30.25±4.61 in Ⅰ, Ⅱstages, 38.42±5.88 in Ⅲstage. Statistic analysis showed that the MVD had siginicantly different in different TNM stages(T=2.173,p=0.038). ⑥The mean of MVD in esophageal squamous cell carcinoma tissue were 37.53±5.21, 29.54±5.12 with or without lymph node metastasis. Statistic analysis showed that the MVD had siginicantly different in esophageal squamous cell carcinoma tissue with or without lymph node metastasis (T=2.099,p=0.045). ⑦The mean of MVD in esophageal squamous cell carcinoma tissue were 37.67±4.55 in ≤5 years, 25.67±3.88 in >5 years. Statistic analysis showed that the MVD had siginicantly different in different lifetime in esophageal squamous cell carcinoma tissue (T=3.155,p=0.004). 4 The relationship between p53 protein and VEGF: Among 19 cases of p53 protein positive expression, 17 cases of VEGF staining showed positive, whereas among 11 cases of no p53 expression, 6 cases of VEGF staining were negative. Statistic analysis showed that the positive expression rate had siginicantly different in esophageal squamous cell carcinoma tissue between p53 protein and VEGF(r=0.47, X2=4.836, p<0.05). 5 The relationship of p53 protein,VEGF, MVD: In the positive expression of p53 protein the mean of MVD was 38.53±4.43, in the negative was 26.36±4.86.
