Background and Objective:Breast cancer is one of the most common malignancies in women in the world, which threathens women's health seriously. Tumor invasion and metastasis is the main cause of death in patients with breast cancer. Tumor metastatsis and tumor lymph node metastasis are the most important ways of distant metastasis. And hematogenous metastatsis is the way of early metastasis. Tumor angiogenesis and the regulation factors is recently the research focus. This subject is to investigate the expression of hypoxia-inducible factor 1 alpha (HIF-1α) and basic fibroblast growth factor(bFGF) in breast cancer and explore the effects of theirs on carcinogenesis, tumor angiogenesis and progression of breast cancer.Methods:The expression of HIF-1αand bFGF was examined in 67 cases of breast cancer specimens, 39 cases of fibroadenoma of breast by immunohistochemical method and the microvascular density(MVD) was counted in specimens marked by CD105 in the vascular endothelial cells. the relationships between HIF-1α,bFGF expression and the pathologically biological features of breast cancer were assessed.Results:(1) The expression of HIF-1αin breast cancer and fibroadenoma of breast:the positive rate of breast cancer group is 80.6%(54/67), that of fibroadenoma of breast group 0.0%(0/39). Expression of HIF-1αprotein was detected in a significantly greater proportion in breast cancer than that in fibroadenoma of breast(P<0.001); But no significantly difference were found between the tumor tissues with lymph node involved than that of lymph node uninvolved(P>0.05). There were no significant correlation among the expression of HIF-1αin breast cancer and tumor size, tumor location, pathologic grade, lymph node metastasis, clinical staging, age, blood type(P>0.05).(2) The expression of bFGF in breast cancer and fibroadenoma of breast:the positive rate of breast cancer group is 73.1%(49/67), that of fibroadenoma of breast group 35.9% (14/39). Expression of bFGF protein was detected in a significantly greater proportion in breast cancer than that in fibroadenoma of breast(P<0.001); But no significantly difference were found between the tumor tissues with lymph node involved than that of lymph node uninvolved(P>0.05). There were no significant correlation among the expression of bFGF in breast cancer and tumor size, tumor location, pathologic grade, lymph node metastasis, clinical staging,age, blood type(P>0.05).(3) CD105 positive vessel density(MVD) was detected in a significantly greater proportion in breast cancer than that in fibroadenoma of breast(P<0.001). MVD was detected in a significantly greater proportion in positive expression group of HIF-1α, bFGF than that in negative expression group of HIF-1α, bFGF in breast cancer(P<0.01). There were significant correlation among MVD in breast cancer and lymph node metastasis, tumor size and age(P<0.01). There were no significant correlation among MVD in breast cancer and tumor location, pathologic grade, clinical staging, blood type(P>0.05).(4) correlation analysis of bFGF,HIF-1αin breast cancer: the positive rate of HIF-1αin bFGF positive group is 85.71%(42/49), the positive rate of HIF-1αin bFGF negative group is 61.1%(11/18); there was obviously positive correlation between HIF-1αand bFGF(r=0.314,P<0.01).(5) correlation analysis between bFGF,HIF-1αand ER,PR,C-erbB-2 in breast cancer: there was no correlation between bFGF and ER, PR, C-erbB-2 in breast cancer, so does HIF-1α(P>0.05). Conclusion:(1) The expression of HIF-1αand bFGF in the breast cancer tissues are both significantly higher than that in the fibroadenoma of breast tissue. there was obviously positive correlation between HIF-1αand bFGF in breast cancer.(2) There was obviously positive correlation among the expression of HIF-1α, bFGF and MVD in breast cancer. The interaction of HIF-1αand bFGF may play an important role in the regulation of angiogenesis of breast cancer.(3) There were significant correlation among MVD and lymph node metastasis, tumor size and age. MVD can be used as an indicator to judge the prognosis of breast cancer.
