About 97 percent bladder carcinoma is transitional cell carcinoma The transitional cell carcinoma ,one of malignant tumor, has strong power to proliferate . And cell apoptosis is the normal dead of cell to keep the number of cell stable. Recently people study the relationship of apoptosis and carcinoma. They discover that the apoptosis cell decreased in carcinoma tissue compared with normal bladder tissue. Cell apoptosis now is the hot top in the carcinoma study , but it is not enough in bladder transitional cell carcinoma.Recently people discover a new couple of immunization component: Fas and FasL , it has intimate between Fas/FasL mediated apoptosis and tumor happen. Fas and FasL become research focus about molecular biology and apoptosis theory . Between eukaryote cell growth differentiate ,carcinomatous change and cell cycle, it have closely relationship. Cell cycle factor include: cyclin, cyclin dependent kinases(CDKs) and cyclin dependent kinase inhibitors (CDKI) . P27 protein is the product of P27 gene, it is a cyclin dependent kinase inhibitors by recently discovered. P27 can restrain cell proliferation, because P27 restrain cell transformation from G1 stage to S stage. Take place and development of tumor have intimate relationship with abnormality expression of P27.ObjectiveTo investigate to Fas /Fasl and P27 gene expression in bladder tumor andthe significance in evaluating the stage and classification . And to get to know the relationship between the expression and the recurrence of bladder tumor.Materials and Methods50 cases of surgically resected bladder transitional cell carcinoma sample, 12 cases of normal bladder tissue were collected in 1998 -2003. Fas/FasL ,P27 protein expression were detected by SP of immunothistochemistry. The SPSS 11. 5 statistical package program was used for all analysis. X2 - test was used.Result1. The expression rate of Fas in bladder carcinoma was 68% ( 34/50) , in normal tissues was 100% (12/12 ). The positive rate of FasL was 74% (37/ 50) , in normal tissues the positive rate were none. The expression of P27 in carcinoma was 58% (29/50) , but the expression in normal bladder was 100% (12/12). There was significant different P <0.05) above all.2. when the expression of Fas and P27 was compared with grade I and grade II - III There were significant differences ( P < 0. 05 ). The expression rate of Fas in grade I was 18/21 ,but in grade H - III was 16/29. And the expression rate of P27 in grade I was 17/21 ,but in grade II - III was 12/29.3. In invasive tumor(T2T3) , the expression rate of Fas was 13/30, it was lower than superficial tumor(T1) , which was 18/20. (P <0. 05) And there were no significant differences ( P > 0. 05 ) when the expression of Fas or P27 was compared with single tumor and double tumor, or was compared with first occur tumor and recurrent tumor.ConclusionThe low level of Fas, high expression of FasL and the unbalance between them are possible to be an important factor in the onset, progression of carcino-... | | Keywords/Search Tags: | Fas/FasL, P27 Protein, Bladder Carcinoma | PDF Full Text Request | Related items |
